Differences in health-related Quality of Life in patients with or without progression on palbociclib therapy. Preliminary results from the Non-Interventional Study PERFORM
Nghiên cứu đặc điểm thực vật và sơ bộ thành phần hoá học của loài khôi (Ardisia gigantifolia Stapf.), họ Đơn nem (Myrsinaceae)
Mục tiêu: Nghiên cứu đặc điểm thực vật và sơ bộ thành phần hóa học của cây khôi (Ardisia gigantifolia Stapf.), họ Đơn nem (Myrnaceae) Đối tượng và phương pháp nghiên cứu: Các mẫu cây khôi thu hái ở Yên Bái năm 2021 và 2022. Nghiên cứu đặc điểm hình thái thực vật tại thực địa và trong phòng thí nghiệm, xác định tên khoa học bằng phương pháp so sánh đặc điểm hình thái, dựa vào khóa phân loại chi Ardisia. Nghiên cứu đặc điểm vi phẫu: rễ, thân, lá được cắt, tẩy, nhuộm tiêu bản theo phương pháp nhuộm kép, bột thân, lá, rễ được lên tiêu bản bằng phương pháp giọt ép. Định tính các nhóm hợp chất chính trong dược liệu bằng các phản ứng hóa học đặc trưng. Xác định hàm lượng polyphenol tổng số bằng phương pháp đo quang. Kết quả: Xây dựng được bộ dữ liệu về hình thái, cấu tạo giải phẫu và đặc điểm bột dược liệu (rễ, thân, lá) của cây khôi. Thành phần hóa học của loài này gồm các nhóm flavonoid, coumarin, tanin, acid hữu cơ, polysacchrarid, chất béo và sterol. Hàm lượng polyphenol toàn phần trong lá khôi thu hái tại Yên Bái vào tháng 5 đạt cao nhất là 4,05%. Kết luận: Đã xây dựng được bộ dữ liệu về hình thái, cấu tạo giải phẫu và đặc điểm bột dược liệu (rễ, thân, lá) của cây khôi và sơ bộ thành phần hóa học của loài này.

The Mycobacterium abscessus drug development pipeline is poorly populated, with particularly few validated target-lead couples to initiate de novo drug discovery. Trimethoprim, an inhibitor of dihydrofolate reductase (DHFR) used for the treatment of a range of bacterial infections, is not active against M. abscessus. Thus, evidence that M. abscessus DHFR is vulnerable to pharmacological intervention with a small molecule inhibitor is lacking. Here, we show that the pyrrolo-quinazoline PQD-1, previously identified as a DHFR inhibitor active against Mycobacterium tuberculosis, exerts whole cell activity against M. abscessus. Enzyme inhibition studies showed that PQD-1, in contrast to trimethoprim, is a potent inhibitor of M. abscessus DHFR and over-expression of DHFR causes resistance to PQD-1, providing biochemical and genetic evidence that DHFR is a vulnerable target and mediates PQD-1's growth inhibitory activity in M. abscessus. As observed in M. tuberculosis, PQD-1 resistant mutations mapped to the folate pathway enzyme thymidylate synthase (TYMS) ThyA. Like trimethoprim in other bacteria, PQD-1 synergizes with the dihydropteroate synthase (DHPS) inhibitor sulfamethoxazole (SMX), offering an opportunity to exploit the successful dual inhibition of the folate pathway and develop similarly potent combinations against M. abscessus. PQD-1 is active against subspecies of M. abscessus and a panel of clinical isolates, providing epidemiological validation of the target-lead couple. Leveraging a series of PQD-1 analogs, we have demonstrated a dynamic structure-activity relationship (SAR). Collectively, the results identify M. abscessus DHFR as an attractive target and PQD-1 as a chemical starting point for the discovery of novel drugs and drug combinations that target the folate pathway in M. abscessus.

Dysfunction of the central nervous system may inflict spastic movement disorder (SMD). Electrical stimuli were identified as promising therapeutic option. Electrical stimulation provided by a 58-electrode full body garment was investigated based on data from regular trial fittings. Data from 72 testees were investigated. Age averages 36.6 (19.8) ys with 44 females. The cohort spans infantile cerebral paresis (CP) (n=29), multiple sclerosis (MS) (n=23) and stroke (n=20). Data were stratified by etiology and an entry BBS Score<45. Effect sizes (Cohen`s d) related BBS, TUG, FGA, 10mWT, WMFT, EQ5D5L and Pain. Significance levels are indicated by *: p<0.05, **: p<0.01, ***: p<0.001, (t): p<0.1: CP: 1.64***, 0.29*, 1.59***, 0.76(t), 1.00***, 0.5*, 1.28***; MS: 1.83***, 0.83***, 1.28**, 1.07***, 0.93*, 1,11**, 0.78*; Stroke: 1.28**, 0.78**, 0.89, 0.92**, 0.71, 1.26*, 0.78*. Multi-site transcutaneous electrical stimulation may increase ambulation related skills in subjects with SMD stemming from CP, MS and stroke. The results indicate effects on static and dynamic balance, fall risk, mobility, upper extremity improvement and an overall increase in health utility and a reduction in spasticity related pain. Effects are immediate as well as sustained. These results may inspire individual trial fittings and inform further controlled trials.
