Evaluation of gene editing in CHO cells using the Cas‐CLOVER system
Recent advances in gene editing technologies have transformed the genetic engineering of Chinese hamster ovary (CHO) hosts, enabling the development of cell lines with improved stability and productivity. In this study, we employed the programmable nuclease (PN) Cas‐CLOVER to precisely target the Glutamine synthetase (GS) locus in CHO cells. A total of 30 unique serum‐free, suspension‐adapted CHO‐K1 candidate host cell lines were subjected to Cas‐CLOVER‐mediated gene editing, generating over one hundred potential GS knockout (GSKO) clones. A subset of the GSKO clones was subsequently validated using three orthogonal methods to confirm complete knockout of the GS gene in 98 clones. Randomly selected GSKO clones were utilized to produce standard monoclonal antibodies. The resulting pools demonstrated enhanced productivity, with up to a 14.5‐fold increase in titer compared to their wild‐type parental hosts. These findings highlight the potential of gene editing approaches to significantly improve recombinant protein production in CHO expression systems, offering valuable insights for biopharmaceutical manufacturing applications.

Creative giftedness gained a foothold in American K-12 schools with the inception of the Marland report. To understand the growth and direction of creative giftedness over a 50-year timeframe from an international basis, we selected 16 academic journals in the field of gifted education from the United States, Europe, Türkiye, Korea, and Oceania and conducted keyword search strings for creative and Marland . Our retrospective study found precursors on creative giftedness rooted in early psychological research (1900–1950); a transitional conceptualization through a global awakening from Guilford’s presidential speech (1950–2000); and new world interconnectedness with a democratization of creativity that contemplates cyber-creativity (2000–2025). Content analysis of 2,564 keyword article titles showed eight main topics, and we analyzed the distribution of work on these themes, over the three time periods. Trends are observed in terms of journal topics and trends over time across the set of journals.

This paper presents an AI-driven design framework for a compact millimeter-wave (mmWave) antenna targeting defense applications in the 13.5 GHz and $17.25-20 \mathrm{GHz}$ bands. A hybrid Genetic Algorithm-Artificial Neural Network (GAANN) approach is used to optimize antenna geometry and performance metrics including gain, bandwidth, and return loss. The ANN model is trained on simulation data from HFSS to predict performance metrics, allowing the GA to efficiently explore the design space without requiring exhaustive simulations. The hybrid approach achieves significant time reduction and improved design accuracy. The optimized antenna demonstrates high gain, directional stability, and robustness suitable for hostile environmental conditions. This framework represents a novel, rapid, and intelligent approach to mmWave antenna design for defense communication.

Initially the dip is at the middle point between the two Gaussian image points when the separation between two point object is larger than the critical limit. As the distance between the points is reduced, the dip reduces in its upward concavity and it just vanishes at a particular separation of the two point objects.

This article reviews the radiologic findings of recreational drug abuse. Patients with drug abuse often present with a spectrum of related multisystem abnormalities that can range from soft tissue or vascular injury via direct needle penetration to septic emboli, abscesses, septic arthritis, infarct, and hemorrhage. After providing a brief overview of popular routes of administration, we emphasize common and uncommon imaging findings of drug abuse in a system-by-system fashion. A case-based review of multisystem complications from recreational drug use is provided. In this first article of a 2-part series, emphasis is placed on the musculoskeletal and neurological systems.
