Ovine Forestomach Matrix Graft Reduces Surgical Dehiscence in Fasciocutaneous Flap-Based Closure of Pilonidal Disease: A Comparative Study
BackgroundFasciocutaneous flap advancement is a newer alternative surgical approach in pilonidal sinus disease (PSD). This comparative study aimed to assess whether the use of an ovine forestomach matrix (OFM) graft implanted beneath a fasciocutaneous flap closure could minimize dehiscence complications in PSD.MethodsPSD patients who received OFM and a fasciocutaneous flap were prospectively enrolled. Findings were compared to retrospective data from control patients who did not receive an OFM graft. Study outcomes were time to heal, incidence of dehiscence, and scar cosmesis. A within-study cost-utility analysis estimated costs and quality-adjusted life-years (QALYs).ResultsA total of 46 patients who received a flap advancement and OFM graft were compared to 48 retrospective control patients who were treated with flap advancement alone. No difference in median healing time was found in the OFM group (8.0 (IQR: 4.2, 15.8) weeks) compared to the control group (8.0 (IQR: 6.0, 15.2) weeks) (p=0.79). The incidence of dehiscence was significantly less in the OFM group (18%) versus the control group (46%) (p=0.005). The probability of dehiscence in the control group was 3.2 times greater compared to the OFM group. In a cost-utility analysis, the OFM group dominated the control group, with higher QALYs (0.24 vs. 0.23) and lower costs ($4,157 vs. $4,219).ConclusionsThe use of OFM graft with a fasciocutaneous flap resulted in a significantly lower incidence of dehiscence and potentially higher quality-of-life and cost savings, while maintaining a midline incision with favorable cosmesis.

Strategies for Modified Release Oral Formulation Development
The treatment of most illnesses, and in particular chronic diseases, requires achieving and then maintaining a drug concentration in the body that is within the therapeutic range. Modified- and targeted-release drug delivery systems are designed to modulate the drug absorption and/or alter the site of drug release in order to meet specific therapeutic needs that cannot be achieved by conventional oral dosage forms. Capsugel Modified and Targeted Release Technology suite can be divided into three categories: controlled-release drug delivery systems, dual-release and fixed-dose combinations, and site-specific drug delivery systems. Capsugel is continuously developing innovative drug delivery platforms with new functionalities. The aim is to provide a large range of solutions to formulate customers' products, with the double objective to improve patient compliance and therapeutic efficiency, and to meet any site-specific or time-specific drug delivery through the gastrointestinal tract.

Special Aspects of On-Board QC within The Outer Limits of The Continental Shelf of The RF Determining Studies in The Arctic Ocean
Summary Since 2010, geological and geophysical studies were carried out in the Arctic Ocean for the outer limits of the continental shelf of Russian Federation determination outside the 200-mile exclusive economic zone. In all expeditions, one of the key research technique was multibeam echo sounding (MBES), as all the main conventional lines can be drawn based on bathymetric data. Also, in the cause of expeditions of the "Arctic" project seismic, UHR & SBP and gravity surveys were conducted. As the data transferred to the UN Commission on the Limits of the Continental Shelf (CLCS) should meet certain standards on-board real-time QC of all obtained data becomes essential. This is necessary for assessment of the data quality and for prompt adjustment of the survey plans. Furthermore, the compliance of the data with all the criteria set forth in the scientific and technical guidelines of the UN Commission on the Limits of the Continental Shelf and with the main objectives of the studies is important. Consequently, it is necessary to swiftly load the data into a GIS project for construction of formula lines.

Read moreA system identification approach for developing model predictive controllers of antibody quality attributes in cell culture processes
As the biopharmaceutical industry evolves to include more diverse protein formats and processes, more robust control of Critical Quality Attributes (CQAs) is needed to maintain processing flexibility without compromising quality. Active control of CQAs has been demonstrated using model predictive control techniques, which allow development of processes which are robust against disturbances associated with raw material variability and other potentially flexible operating conditions. Wide adoption of model predictive control in biopharmaceutical cell culture processes has been hampered, however, in part due to the large amount of data and expertise required to make a predictive model of controlled CQAs, a requirement for model predictive control. Here we developed a highly automated, perfusion apparatus to systematically and efficiently generate predictive models using application of system identification approaches. We successfully created a predictive model of %galactosylation using data obtained by manipulating galactose concentration in the perfusion apparatus in serialized step change experiments. We then demonstrated the use of the model in a model predictive controller in a simulated control scenario to successfully achieve a %galactosylation set point in a simulated fed‐batch culture. The automated model identification approach demonstrated here can potentially be generalized to many CQAs, and could be a more efficient, faster, and highly automated alternative to batch experiments for developing predictive models in cell culture processes, and allow the wider adoption of model predictive control in biopharmaceutical processes. © 2017 The Authors Biotechnology Progress published by Wiley Periodicals, Inc. on behalf of American Institute of Chemical Engineers Biotechnol.
