ABSTRACTSecreted proteins mediate intercellular communication throughout the lives of multicellular organisms. However, due to the lack of new technology for secreted protein capturing, the progress of “secretomics’’ lags behind. Here, we report a two-step secretome enrichment method (tsSEM) combining unnatural amino acid labeling and click chemistry-based biorthogonal reaction, which enablesin vitrosecretome profiling in the presence of serum. Using this novel method, we systematically investigated the secretome of human iPSCs-derived astrocytes (iAst) in different disease models and identified a panel of astrocyte-secreted proteins that are responsible for its non-cell autonomous toxicity under disease conditions. Furthermore, we validated two astrocytes-derived novel neurotrophic proteins, FAM3C and KITLG, which we identified from disease models, and found that they could boost neurite outgrowth, protect neurons, and promote neural progenitor proliferation. Our study highlights the utility of secretome profiling of iAst and demonstrates its applications in disease study and target identification and validation in drug development.

The Vera C. Rubin Observatory is reaching the final stages of its construction and integration, advancing towards its 10-year Legacy Survey of Space and Time (LSST). One of the key milestones was the installation of the M1M3 Mirror Cell Assembly onto the Simonyi Survey Telescope’s (SST) Telescope Mount Assembly (TMA). The Cell Assembly actively supports the primary/tertiary mirror (M1M3), playing a crucial role in maintaining the glass safe and ensuring image quality. However, before the mirror glass installation, the Cell Assembly was installed on the TMA while supporting a steel surrogate M1M3 mirror. This surrogate closely mimics the glass mirror’s mass, center of gravity, and geometry. The M1M3 cell and surrogate were tested under conditions that simulate rapid field changes in the sky, which are essential for the observatory’s ambitious sky mapping schedule. These tests, extending from 1-100% of designed telescope slew velocities/accelerations, assessed the M1M3 active mirror support system, including the force balance system’s performance, the hardpoint behaviors, and the efficacy of the pneumatic figure control actuators. Preliminary results suggest the system meets operational requirements, ensuring safety and effectiveness at full speed.
Read moreCharge Dynamics Engineering Sparks Hetero‐Interfacial Polarization for an Ultra‐Efficient Microwave Absorber with Mechanical Robustness (Small 6/2024)
Interfacial Charge Dynamics In article number 2306104, Xiaolong Jia and co-workers clarified the charge dynamics mediated by the work function within the heterointerface. This understanding guides the proposal of a surface chemistry-tailored strategy aimed at sparking hetero-interfacial polarization between carbon fibers and ZnO nanowires. Consequently, the microwave attenuation ability is synergistically aligned with interfacial bonding, bestowing polymer-based composites with remarkable microwave absorption efficiency and mechanical robustness.

Abstract In order to solve the short stabilization time of the water‐in‐oil emulsion binder system and the shortcomings of the critical blast size of the explosive ink molding sample, a water‐in‐oil emulsion binder was designed with polyvinyl alcohol (PVA) aqueous solution as the aqueous phase, fluorocarbon resin (FEVE) ethyl acetate solution as the oil phase, and toluene diisocyanate (TDI) added to modify the water‐in‐oil emulsion binder. The hexanitrohexaazaisowurtzitane(CL‐20)based explosive ink was made by incorporating submicron CL‐20 into the binder system, and the print‐formed samples were tested for microscopic morphology, crystallographic analysis, mechanical susceptibility, and detonation transmission performance. The results showed that the explosive ink‘s viscosity was moderate and suitable for direct printing, and the formed sample‘s surface particles were uniformly dispersed and had a honeycomb structure. The crystal form of CL‐20 explosive in the explosive ink did not change, the molded sample‘s impact and frictional sensitivities were 1 J and 32 N lower than those of submicron CL‐20, respectively, and the detonation velocity of the molded sample was 6655 m/s. The critical detonation size and critical detonation thickness were 1×0.0078 mm and 0.1 mm, respectively, and a critical detonation corner turning of 90°–160° could be achieved. The results show that the prepared binder system is stable for a long time, and the explosive ink molding samples have excellent blast transmission performance.

Unilateral nevoid telangiectasia (UNT) is a benign, rare dermatosis, which primarily involves the face and neck on the trigeminal and cervical dermatomes (third and fourth). It is characterized by telangiectatic, unilateral vascular lesions of linear, segmental or metameric distribution. It may be congenital or acquired, such as in the case of physiological or pathological estrogen excess presented in puberty, pregnancy or chronic liver disease. Such condition is related to an increase of estrogen and progesterone receptors in the involved skin. A 22-year-old male patient presents with dermatosis on the right thigh’s inner face, clinically and histologically compatible with UNT of rare localization.

In order to fully identify the root cause of cryogenic pneumatic valve failure in the process of liquid rocket engine test, and to analyze the dynamic time change of the process of cryogenic pneumatic valve reliability analysis, this paper first analyzes the failure factors of low temperature pneumatic valve in the test process of liquid rocket engine according to the human-machine environment system engineering. By introducing the dynamic Bayesian network theory, and by means of analyzing the time-change failure failure mode of cryogenic pneumatic valve, constructing the time-change reliability model of cryogenic pneumatic valve was constructed in the test process of liquid rocket engine, and the reliability growth technology of cryogenic pneumatic valve was researched by the optimal failure search strategy. Finally, the validity and accuracy of this method are verified by the example. This method can provide a method guide for quantitative analysis and improvement of the reliability of cryogenic pneumatic valve, and provide support for the accurate and objective evaluation and development of liquid rocket engine performance.
Read moreElimination-based certificates for triangular equivalence and rank profiles
In this paper, we give novel certificates for triangular equivalence and rank profiles. These certificates enable somebody to verify the row or column rank profiles or the whole rank profile matrix faster than recomputing them, with a negligible overall overhead. We first provide quadratic time and space non-interactive certificates saving the logarithmic factors of previously known ones. Then we propose interactive certificates for the same problems whose Monte Carlo verification complexity requires a small constant number of matrix-vector multiplications, a linear space, and a linear number of extra field operations , with a linear number of interactions. As an application we also give an interactive protocol, certifying the determinant or the signature of dense matrices, faster for the Prover than the best previously known one. Finally we give linear space and constant round certificates for the row or column rank profiles.
