The growing resistance of Enterobacterales to last-resort antibiotics such as carbapenems puts a significant burden on healthcare systems, also due to plasmids driving a rapid spread of carbapenem resistance. We here evaluate the use of long-read nanopore sequencing to investigate carbapenem resistance dynamics and the role of plasmid transfers and environmental reservoirs in the hospital setting. Over 13 months, routine clinical diagnostics identified recurring isolates of carbapenem-resistant Citrobacter species carrying Klebsiella pneumoniae carbapenemases (KPCs) and/or OXA-48-like carbapenemases from patient screening and hospital drain samples. While routine diagnostic approaches provided limited insights into the carbapenem resistance dynamics, we show that near-complete de novo assembly of chromosomes and plasmids by long-read nanopore sequencing allowed for high-resolution strain identification, plasmid profiling, and antibiotic resistance gene detection. Notably, genomically nearly indistinguishable Citrobacter freundii of the high-risk sequence type ST91 genomes were recovered from screening samples collected in the same hospital room 1 year apart. We further provide evidence of a KPC-2-encoding IncN plasmid that is likely to have spread across bacterial species and between patient and drain isolates, which emphasizes the role of contaminated drains in the persistence and dissemination of antimicrobial resistance within the hospital environment. Overall, this study demonstrates the value of long-read nanopore sequencing for uncovering the complex dynamics of carbapenem resistance spread and persistence in the hospital setting and its potential implications for infection prevention and control.

In the context of Site Reliability Engineering (SRE), predicting server-side anomalies is critical for maintaining web service availability. This study addresses the prediction of anomalous HTTP status codes using machine learning on a real-world dataset from an E-commerce site. We monitored eight predictors, with anomalous codes representing only 1% of the data. Three gradient boosting models (Categorical Boosting (CatBoost), Extreme Gradient Boosting (XGBoost), Light Gradient Boosting Machine (LightGBM)) were tuned with Optuna, and class imbalance was mitigated using resampling methods (Condensed Nearest Neighbors (CNN), Random Undersampling (RUS), Cluster Centroids (CC), Synthetic Minority Oversampling Technique (SMOTE)). Evaluation yielded precision, recall, and F1-scores of 0.94, 0.98, and 0.97, respectively. SMOTE achieved the best results, demonstrating its applicability for rare Anomaly Detection (AD).

Understanding detailed energy consumption patterns is crucial for optimizing resource allocation in microgrids, which often incorporate renewable energy sources and stationary batteries. In this context, this paper addresses the Non-intrusive Load Monitoring problem of appliance-state classification, providing insights into appliance-specific usage by using only the aggregate active power consumed in a building. Deep Neural Networks represent the state of the art in this field, but they require a significant amount of data for training. Consequently, in recent years, the research community has explored weakly supervised approaches. Although weakly supervised methods have proven effective, they rely on the use of pooling functions to calculate bag-level predictions from instance-level ones. These functions should be intrinsically related to the dynamics of the input signal, and while several alternatives have been proposed in the literature, none have specifically addressed the characteristics of appliance power profiles. This paper fills this gap by introducing a new pooling function, Contextual Pooling, that operates on multiple adjacent instance-level predictions to better capture the dynamics of appliance activations. This approach has been evaluated against six different methods on the UK-DALE and REFIT datasets. The results indicate that, on average, Contextual Pooling improves performance by at least 0.9 percentage points on UK-DALE and by 0.7 percentage points on REFIT compared to benchmark methods. McNemar’s test confirms the statistical significance of these improvements.

This study explores the cost-benefit elements of construction waste management (CWM) within China's rapidly expanding construction sector. As urbanization accelerates, the generation of construction waste has surged, posing significant environmental and economic challenges. This research analyzes various management strategies, including waste reduction, recycling, and sustainable disposal methods, to assess their economic viability and environmental impact. Through a combination of qualitative studies, the study identifies cost-benefit elements. The findings underscore the importance of integrating effective waste management practices in construction projects, highlighting potential cost savings and environmental benefits. This research aims to provide actionable insights for policymakers and industry stakeholders, promoting a more sustainable construction paradigm in China.
