AI in Drug Discovery and Development

Drug discovery has traditionally been a time consuming and expensive endeavor. Additionally, drugs weren’t as effectively designed as those that are being predicted and developed through AI and ML today. Machine learning is a form of artificial intelligence that develops and evolves based on experience (similarly to the human mind), and is more recently being utilized in drug discovery and design. The integration of AI and ML into the drug discovery and development process has allowed for higher target precision, lower toxicity, and better dosage formulations. AI more generally has been introduced to and has been leveraged at, each step of drug development, including target identification and validation, hit identification, as well as hit to lead optimization, and has been key in shortening the previously lengthy drug screening process. AI and ML has also been applied downstream in drug formulation where it has maximized resource utilization and is allowing for web-based 3D printing of drugs. Application of AI in the drug development process has also been extended to the modeling of novel drug-like compounds to predict their ADMET properties. This review will address the stages of drug discovery and development in which the application of AI and ML modeling has altered the traditional development of drugs. The drug discovery process has been time-consuming and expensive in the past. Also, previous drugs were not formulated as well as the drugs being expected and developed using AI and ML technologies. This article shall elaborate on the stages of drug discovery and development where AI and ML modelling have revolutionized the traditional methods of drug development. AI, in the broader context, has been taken up and implemented at each stage of drug discovery, from target identification and validation to hit finding and the progression from hit to lead optimization, with a vital role in streamlining the formerly time-consuming process of drug screening. Several methods based on machine learning are being used to predict drug targets, predict the structures of drug targets, predict binding sites, perform ligand-based similarity searches, design ligands with certain desired properties de novo, develop scoring functions for molecular docking, build Quantitative structure-activity relationship (QSAR) models for the prediction of biological activity, and predict the pharmacokinetic and pharmacodynamic properties of ligands.

Artificial intelligence: Machine learning approach for screening large database and drug discovery

Artificial Intelligence (AI) is revolutionizing the pharmaceutical landscape by transforming the traditionally laborious, expensive, and time-consuming drug discovery and development process. By harnessing the power of machine learning (ML), deep learning (DL), and advanced data analytics, AI systems enable rapid screening of compounds, prediction of drug-target interactions, and optimization of molecular structures. These technologies significantly reduce development timelines and costs while increasing the probability of success in clinical trials. AI-driven platforms facilitate personalized medicine through the integration of pharmacogenomic data, allowing for tailored treatments based on individual patient profiles. Moreover, AI contributes to critical areas such as virtual screening, dosage prediction, protein-protein interaction modeling, and drug repurposing. Despite challenges related to data quality, ethical considerations, and regulatory compliance, AI continues to prove its potential in streamlining end-to-end drug development. As the industry moves toward more intelligent and data-driven frameworks, AI stands as a pivotal force in accelerating therapeutic innovation and improving global health outcomes. Drug design and development are crucial areas of study for chemists and pharmaceutical companies. Nevertheless, the significant expenses, lengthy process, inaccurate delivery, and limited effectiveness present obstacles and barriers that affect the development and exploration of new drugs. Moreover, big and complex datasets from clinical trials, genomics, proteomics, and microarray data also disrupt the drug discovery approach. The integration of Artificial Intelligence (AI) into drug design is both timely and crucial due to several pressing challenges in the pharmaceutical industry, including the escalating costs of drug development, high failure rates in clinical trials, and the increasing complexity of disease biology. AI offers innovative solutions to address these challenges, promising to improve the efficiency, precision, and success rates of drug discovery and development. Artificial intelligence (AI) and machine learning (ML) technology are crucial tools in the field of drug discovery and development. More precisely, the field has been revolutionized by the utilization of deep learning (DL) techniques and artificial neural networks (ANNs). DL algorithms & ML have been employed in drug design using various approaches such as physiochemical activity, polypharmacology, drug repositioning, quantitative structure-activity relationship, pharmacophore modeling, drug monitoring and release, toxicity prediction, ligand-based virtual screening, structure-based virtual screening, and peptide synthesis. The use of DL and AI in this field is supported by historical evidence. Furthermore, management strategies, curation, and unconventional data mining aided assistance in modern modeling algorithms. In summary, the progress made in artificial intelligence and deep learning algorithms offers a promising opportunity for the development and discovery of effective drugs, ultimately leading to significant benefits for humanity. In this review, several tools and algorithmic programs have been discussed which are being used in drug design along with the descriptions of the patents that have been granted for the use of AI in this field, which constitutes the main focus of this review and differentiates it fromalready published materials.

In pharmaceutical research drug discovery and development process is timeconsuming and expensive. In many cases, it produces incompetent results due to the failure of in vitro and in vivo conventional approaches. Before any new drug is placed in the market it must undergo rigorous testing to get FDA approval. Due to the several limitations imposed by the drug discovery process, in recent times in silico approaches are widely applied in this field. The purpose of this review is to highlight the current molecular docking strategies used in drug discovery and to explore various advances in the field. In this review we have compiled database after an extensive literature search on docking studies which has found its applications relevant to the field of formulation and development. The papers retrieved were further screened to appraise the quality of work. In depth strategic analysis was carried out to confirm the credibility of the findings. The papers included in this review highlight the promising role of docking studies to overcome the challenges in formulation and development by emphasizing it's applications to predict drug excipient interactions which in turn assist to increase protein stability; to determine enzyme peptide interactions which maybe further used in drug development studies; to determine the most stable drug inclusion complex; to analyze structure at molecular level that ascertain an increase in solubility, dissolution and in turn the bioavailability of the drug; to design a dosage form that amplify the drug discovery and development process. This review summarizes recent findings of critical role played by molecular docking in the process of drug discovery and development. The application of docking approach will assist to design a dosage form in the most cost effective and time saving manner.

Computer-aided drug discovery and development (CADDD): In silico-chemico-biological approach

Introduction: Contract Research Organizations (CROs), Clinical Research Centers/Networks and private research institutions play an important role in the clinical research process, especially in the drug discovery and development process. They act as essential partners for pharmaceutical drug delivery/development and biotech companies. Over the last few decades, the demand for outsourcing of clinical research by pharmaceutical companies has increased, with the market for CRO and private research institutional services growing significantly. This trend has been driven by a need for increased productivity and efficiency to meet the increasing research and development costs, as well as the complexities associated with modern drug development. Statement of objective: This study has an objective to establish CROs with known recognition in sub Saharan Africa. Special emphasis on Cameroon institutions regulating and conduct clinical research. Methodology: Historically, drug discovery services were predominantly conducted in-house, safeguarding intellectual property and close collaboration with internal teams. However, the competitive nature of the sector has motivated pharmaceutical companies to adopt an outsourcing model, allowing them to decentralize, outsourced and exploit specialized expertise and technologies offered by the CROs. These partnerships enable drug developers to accelerate the identification and validation of promising drug candidates through streamlined pre-clinical and clinical trial processes. This is systematic research findings that we conducted a systematic search of the source documents from organizations, access to websites and google search to assemble our information, e-data information system. The systematic search gave us information on the contract organizations with online visibility. Vital information from annual reports, web information system management of the different organizations were accessible for easy navigations. Information on laws governing clinical research and major regulators in Cameroon. Results: Through a systematic online search contract research organization operating in Cameroon and Sub Saharan Africa that were available and accessible on line were identified. The list was not exhaustive as some CROs may be in existence but we could not have access to their activities and home page for enough visibility online. The organizations and CROs operating in Cameroon have been well illustrated. The laws and regulations governing clinical research in Cameroon has been well illustrated. It was observed that CROs face multiple challenges. The Contract Research Organization (CRO) are faced with multiple challenges that can be a limiting factor to their growth and quality service delivery: These challenges are listed though not exhaustive as Technological Disruption, Regulatory landscape, intellectual Property Issues, Economic Volatility, Limited Human resource expertise, Return on investment/High Costs: Conclusion/Recommendation: The CROs in Cameroon are required to be in compliance with the law regulating research in human subjects of 2022 and the regulations on Clinical research in Cameroon. In sub Saharan Africa CROs are aligned with the regulation enforced in their different countries. The success of CROs is based on their compliance to innovative trends in clinical research and the capacity to strengthen collaboration among policy makers and actors. The ability to face challenging opportunities in clinical research and embrace transformative breakthroughs that can bring faster new chemical entities to the market at the same time improving the standard of care (SOC) of the population. In the future, the synergy between innovation, strategic partnerships, and a commitment to excellence for CROs shall be the cornerstones of future success in conducting clinical research for Pharmaceutical drug discovery and development Based on the regulatory demanding nature of clinical regulation in Cameroon CROs must have a legal jurisdiction and recognition of their operation, and conform to the norms and standard laid down by regulatory authorities and the states of their jurisdiction CROs offer a wide range of services to support the development and execution of clinical trials, ensuring compliance with regulatory requirements and ethical standards. Clinical Research Organizations (CROs) play a pivotal role in the pharmaceutical, biotechnology, and medical device industries as a research service delivery provider on a contract basis.

The development of sound bioanalytical method(s) is of paramount importance during the process of drug discovery and development culminating in a marketing approval. Although the bioanalytical procedure(s) originally developed during the discovery stage may not necessarily be fit to support the drug development scenario, they may be suitably modified and validated, as deemed necessary. Several reviews have appeared over the years describing analytical approaches including various techniques, detection systems, automation tools that are available for an effective separation, enhanced selectivity and sensitivity for quantitation of many analytes. The intention of this review is to cover various key areas where analytical method development becomes necessary during different stages of drug discovery research and development process. The key areas covered in this article with relevant case studies include: (a) simultaneous assay for parent compound and metabolites that are purported to display pharmacological activity; (b) bioanalytical procedures for determination of multiple drugs in combating a disease; (c) analytical measurement of chirality aspects in the pharmacokinetics, metabolism and biotransformation investigations; (d) drug monitoring for therapeutic benefits and/or occupational hazard; (e) analysis of drugs from complex and/or less frequently used matrices; (f) analytical determination during in vitro experiments (metabolism and permeability related) and in situ intestinal perfusion experiments; (g) determination of a major metabolite as a surrogate for the parent molecule; (h) analytical approaches for universal determination of CYP450 probe substrates and metabolites; (i) analytical applicability to prodrug evaluations-simultaneous determination of prodrug, parent and metabolites; (j) quantitative determination of parent compound and/or phase II metabolite(s) via direct or indirect approaches; (k) applicability in analysis of multiple compounds in select disease areas and/or in clinically important drug-drug interaction studies. A tabular representation of select examples of analysis is provided covering areas of separation conditions, validation aspects and applicable conclusion. A limited discussion is provided on relevant aspects of the need for developing bioanalytical procedures for speedy drug discovery and development. Additionally, some key elements such as internal standard selection, likely issues of mass detection, matrix effect, chiral aspects etc. are provided for consideration during method development.

The gauntlet for a new drug to succeed is a daunting one. The reduction in the time and cost required for modern drug discovery and development serves as a crucial need. An important element to accelerate drug discovery and development process is the rapid identification of promising drug candidates as opposed to the non-starters before unnecessary vast sums are invested. The aim of molecular imaging is to precisely visualize, characterize, and measure biological processes at the molecular and cellular levels in humans and other living systems [1]. By introducing molecular imaging probes into traditional diagnostic imaging techniques, researchers can determine the expression of indicative molecular markers at different stages of diseases. The introduction of new imaging probes, methods, and advanced imaging instrumentation is significantly speeding up the processes of drug discovery and development. The convergence of innovations has created more sensitive, specific and higher resolution measurements within living organism, especially small animals. This is expected to improve a wide range of discovery activities such as target biology, compound screening, pharmacokinetics (PK), and pharmacodynamics (PD) evaluation in animal disease models and, eventually, clinical trials. In this chapter, we will summarize and evaluate the applications of various advanced small animal imaging techniques in drug development process.

ABSTRACTThe most strategic weapon in drug discovery in the recent past has been artificial intelligence (AI)—bringing new approaches to one of the toughest areas of the pharmaceutical industry. Various AI approaches such as DL and ML methods utilized in various stages of drug discovery and development including but not limited to virtual screening and target identification are also discussed here. Employing this approach, this review looks at AI programs and platforms that exist in drug discovery today in a bid to outline what a future with AI in this field has in stock. In addition to this, this review does not only give a momentary state of the state of affairs of the AI in the space, but also briefly discusses what is in store next, along with the drawback and the opportunity more so from this perspective.

Drug discovery is a multifaceted and resource-intensive process traditionally marked by long development timelines, high financial costs, and significant attrition rates. The integration of Artificial Intelligence (AI) has introduced transformative opportunities to overcome these barriers by enhancing data-driven decision-making, accelerating molecular design, and improving clinical translation. This chapter explores the foundations, applications, and case studies of AI across all stages of drug discovery - from target identification and validation to lead optimization and clinical development. Real-world examples, including BenevolentAI, Atomwise, Insilico Medicine, and the Pfizer–IBM Watson collaboration, illustrate AI’s role in accelerating drug design, repurposing, and optimization. Despite its promise, challenges such as data quality, model interpretability, regulatory uncertainty, and intellectual property disputes continue to impede widespread adoption. The chapter concludes by emphasizing the future directions of AI in synergy with quantum computing, multi-omics integration, and personalized medicine. Overall, AI represents a paradigm shift in pharmaceutical innovation, poised to make drug discovery faster, more precise, and more cost-effective while promoting a human–AI collaborative ecosystem for sustainable biomedical advancement.

Abstract: The drug discovery landscape and neurodegenerative disease diagnostics are expe-riencing a paradigm shift with the integration of artificial intelligence (AI) and machine learn-ing (ML) technologies. This review provides an elaborate discussion of AI-based approaches across all stages of computer-aided drug design (CADD), including virtual screening, peptide synthesis, pharmacophore modelling, quantitative structure–activity relationships (QSAR), and drug repurposing. Particular emphasis is placed on Alzheimer's disease (AD), a multifac-eted neurodegenerative disorder, with AI enabling early diagnosis, patient stratification, and subtype classification to support precision medicine. The review highlights recent developments in supervised and unsupervised learning, omics integration, and the increasing importance of explainable AI (XAI) in addressing the “black box” limitations of traditional AI models. We categorize and assess major AI platforms and tools by their scope, stage of implementation, and form of explainability, offering a practical framework for their application in pharmaceutical and clinical research. The combined use of deep learning, quantum simulation, and virtual reality is also discussed in the context of de novo drug design, compound screening, and toxicity prediction. With con-tinued advances in AI, interdisciplinary collaboration and ethical oversight are essential to translate these digital innovations into effective, safe, and personalized therapies. This review serves as an all-encompassing guide for researchers, clinicians, and developers working at the interface of AI and drug development, particularly those addressing the unmet challenges in Alzheimer's disease. As outlined in the abstract and reflected in the article title, this review bridges AI-driven drug discovery and Alzheimer’s diagnosis, with specific empha-sis on explainable models for next-generation therapeutics. The subsequent sections expand on these themes, offering a structured synthesis of tools, applications, and methodological frameworks.
