Abstract

Cell-based therapy (CBT) has garnered significant interest as a potential treatment for incurable diseases. In CBT, the cells are the final products and require stringent processes in terms of safely delivering them to patients. This paper discusses new technology that is being developed to allow for real-time monitoring of cell production quality during the biomanufacturing of cell therapies. The aim is to develop a novel switchable biosensor for applications in continuous cell therapy process monitoring. The biosensors incorporate stimuli-responsive technology that allows for the biosensor to be switched on or off electrically for on-demand sensing of cytokines. In this preliminary work, we show the coupling of nanobodies targeted to eGFP via carbodiimide chemistry to immobilize them onto 11-mercaptoundecanoic acid gold screen-printed electrodes (SPE). The gold chips were characterized using spectroscopic ellipsometry and gold SPEs were characterized with cyclic voltammetry and faradaic electrical impedance spectroscopy (EIS). Our results show successful immobilization of GFP nanobodies onto the SPE biosensors and detection of eGFP through EIS and fluorescence microscopy.
