Image plane adjustment in photoacoustic tomography based on a full ring transducer array

Compared to piezoelectric based photoacoustic (PA) scanners, the planar Fabry-Perot (FP) scanner has several advantages. It can provide small element size with high sensitivity, a smooth broadband frequency response, and is transparent to excitation light. This enables the FP scanner to provide excellent high-resolution in vivo PA images of soft tissue to depths up to approximately 10 mm. However, unlike piezoelectric scanners, the FP scanner in its current form cannot provide a pulse-echo ultrasound (US) as well as a PA image, which is useful because of the additional tissue contrast it provides. To address this, a dual mode FP scanner-based system that, for the first time, can acquire co-registered 3D PA and US images has been developed. In order to provide an optical US generation capability, the FP ultrasound sensor was coated with a novel Gold-Nanoparticle-PDMS composite which was excited with nanosecond laser pulses to generate plane wave US pulses. By modifying the FP sensor in this way, it now acts as an US transmitter as well as a receiver. The coating is highly absorbing at the US generation wavelength (>95%) but transparent at the PA excitation wavelength, the latter to allow the system to also operate in PA imaging mode as before. The generated US pulses exhibited peak pressures in the MPa range, which is comparable to the output of conventional piezoelectric based medical US scanners. The pulses had a broad bandwidth (>40 MHz) and the emitted wavefront was planar to within λ/10 at 10 MHz. PA and pulse-echo US signals were mapped in turn by the FP scanner over centimetre scale areas with a step size of 100 μm and an element size of 64 μm. The -3dB bandwidth of the FP sensor was 30 MHz. Reconstruction methods using a k-space formulation recovered co-registered 3D PA and US images. The system’s lateral spatial resolution was evaluated by imaging a line target at depths up to 10 mm and ranged between 50 and 120 μm for both modes. Arbitrarily shaped 3D objects were imaged to demonstrate the volumetric US imaging capability of the scanner. Tissue mimicking phantoms, with impedance mismatches representative of soft tissues, and ex vivo tissue samples were imaged with the system as well as a conventional clinical US scanner for comparison. Finally, the system obtained promising high-resolution 3D dual mode PA-US images for a variety of phantoms with contrast based on both optical absorption and acoustic impedance. This novel all-optical system has the potential to add complementary morphological contrast to photoacoustic vascular images which could aid the clinical assessment of superficial tumours, lymph node disease and other conditions.

Photoacoustic imaging (PAI) used for noninvasive imaging of biological tissue has been reported in many literature. However, there are still some disadvantages in the novel technique, such as the poor efficiency of imaging. In the current PAI, multiple excitation of laser and multiple acquisition of signal are necessary for image reconstruction. In this case, laser pulses may injure biological tissue due to energy accumulation. To popularize PAI in clinical applications, it is necessary to develop a new imaging approach to increase the efficiency of PAI. A spatial phase-controlled algorithm is presented for full-field three-dimensional (3D) image reconstruction. By using the algorithm, photoabsorption sources at different depths can be reconstructed using just one set of data acquired in single laser shot. Unfocused plane transducer array and parallel data-acquisition (PDA) equipment are used for real-time photoacoustic (PA) signal detection and acquisition. The spatial resolution of the 3D PAI system was analyzed. Two graphite rods at various positions in a simulation model and two bifurcate vessels in the ear of rabbit were imaged. In addition, the motion trace of one particle flowing at constant velocity was captured dynamically. Experimental results showed that spatial phase-controlled algorithm based on plane transducer array and PDA system was capable of static and dynamic 3D PAI. Spatial phase-controlled algorithm is introduced for 3D image reconstruction. The PA signals are collected by plane transducer array and PDA system in single pulse excitation. The acquired volumetric data are sufficient for 3D image reconstruction. Therefore, tissue can avoid the long-term exposure to light source and it is safer than the current PAI for in vivo imaging. With an increase in the repetition rate of laser pulse and speed of image display, the imaging method will realize real-time 3D imaging, which will be significant in clinical detection and medical diagnosis.

An inexpensive noncontact photoacoustic (PA) imaging system using a low-power continuous wave laser and a kilohertz-range microphone has been developed. The system operates in both optical and PA imaging modes and is designed to be compatible with conventional optical microscopes. Aqueous coupling fluids are not required for the detection of the PA signals; air is used as the coupling medium. The main component of the PA system is a custom designed PA imaging sensor that consists of an air-filled sample chamber and a resonator chamber that isolates a standard kilohertz frequency microphone from the input laser. A sample to be examined is placed on the glass substrate inside the chamber. A laser focused to a small spot by a 40 × objective onto the substrate enables generation of PA signals from the sample. Raster scanning the laser over the sample with micrometer-sized steps enables high-resolution PA images to be generated. A lateral resolution of 1.37 ?? ? m was achieved in this proof of concept study, which can be further improved using a higher numerical aperture objective. The application of the system was investigated on a red blood cell, with a noise-equivalent detection sensitivity of 43,887 hemoglobin molecules ( 72.88 × 10 ? 21 ?? mol or 72.88 zeptomol). The minimum pressure detectable limit of the system was 19.1 ?? ? Pa . This inexpensive, compact noncontact PA sensor is easily integrated with existing commercial optical microscopes, enabling optical and PA imaging of the same sample. Applications include forensic measurements, blood coagulation tests, and monitoring the penetration of drugs into human membrane.

Photoacoustic (PA) imaging is an emerging technology that could map the functional contrasts in deep biological tissues in high resolution by “listening” to the laser induced thermoelastic waves. Almost all of the current studies in PA imaging are focused on the intensity of the PA signals as an indication of the optical absorbance of the biological tissues. Our group has for the first time demonstrated that the frequency domain power distribution of the broadband PA signals encode the texture information within the regions-of-interest (ROI). Following the similar method of ultrasound spectral analysis (USSA), photoacoustic spectrum analysis (PASA) could evaluate the relative concentrations and, more importantly, the dimensions of microstructures of the optically absorbing materials in biological tissues, including lipid, collagen, water and hemoglobin. By providing valuable insights into tissue pathology, PASA should benefit basic research and clinical management of many diseases, and may help achieve eventual “noninvasive biopsy”. In this work, taking advantage of the optical absorption contrasts contributed by lipid and hemoglobin at 1200-nm and 532-nm wavelengths respectively, we investigated the capability of PASA in identifying histological changes corresponding to fat accumulation livers through the study on ex vivo and in situ mouse models. The PA signals from the mouse livers were acquired using our PA and US dual-modality imaging system, and analyzed in the frequency domain. After quantifying the power spectrum by fitting it to a first order model, three spectral parameters, including the intercept, the midband fit and the slope, were extracted and used to differentiate fatty livers from normal livers. The comparison between the PASA parameters from the normal and the fatty livers supports our hypotheses that PASA can quantitatively identify the microstructure changes in liver tissues for differentiating normal and fatty livers.

Photoacoustic (PA) imaging technology is expected to be applied to clinical assessment for peripheral vascularity. We started a clinical evaluation with the prototype PA imaging system we recently developed. Prototype PA imaging system was composed with in-house Q-switched Alexandrite laser system which emits short-pulsed laser with 750 nm wavelength, handheld ultrasound transducer where illumination optics were integrated and signal processing for PA image reconstruction implemented in the clinical ultrasound (US) system. For the purpose of quantitative assessment of PA images, an image analyzing function has been developed and applied to clinical PA images. In this analyzing function, vascularity derived from PA signal intensity ranged for prescribed threshold was defined as a numerical index of vessel fulfillment and calculated for the prescribed region of interest (ROI). Skin surface was automatically detected by utilizing B-mode image acquired simultaneously with PA image. Skinsurface position is utilized to place the ROI objectively while avoiding unwanted signals such as artifacts which were imposed due to melanin pigment in the epidermal layer which absorbs laser emission and generates strong PA signals. Multiple images were available to support the scanned image set for 3D viewing. PA images for several fingers of patients with systemic sclerosis (SSc) were quantitatively assessed. Since the artifact region is trimmed off in PA images, the visibility of vessels with rather low PA signal intensity on the 3D projection image was enhanced and the reliability of the quantitative analysis was improved.

Photoacoustic (PA) imaging is a hybrid imaging technology which combines the best of optical (contrast) and ultrasound (resolution) imaging. PA imaging uses intrinsic contrast agents in the body like blood (hemoglobin), melanin, etc. But the contrast from the intrinsic contrast agents might not be sufficient for different applications. So, external contrast agents are needed for improving the contrast of PA images. Organic dyes, inorganic dyes, and nanomaterials can be used as photoacoustic contrast agents. The major issue with using external contrast agent is that they often need FDA approval to be used for in-vivo studies. Availability of FDA approved contrast agents for PA imaging is very limited. In this work, we present the feasibility of using food and food-based dyes as photoacoustic contrast agents. We use commonly used foods like coffee, tea, chocolate and food colorants as contrast agents. We use alpinion’s E-CUBE dual mode ultrasound and photoacoustic imaging system with mobile Nd:YAG laser pumped by OPO laser to demonstrate the efficiency of the contrast agents. The contrast agents were compared with methylene blue. Out of the many different agents we tested, coffee, chocolate and few other dyes proved as efficient photoacoustic contrast agents. We performed photoacoustic spectroscopy to identify at which wavelength the dye performed best. We also tested the contrast agents for imaging sentinel lymph nodes in rats and the results are very similar to methylene blue. This will enable the transition of photoacoustic imaging to the clinics more easily.

Photoacoustic imaging is a powerful tool for assessing tumor vasculature and oxygen saturation as well as detecting contrast agents for molecular imaging. Another exciting possibility is to visualize gene expression by using a reporter which produces a photoacoustic contrast agent. Tyrosinase is such a reporter in that its expression results in the production of melanin, which gives a strong photoacoustic signal. Here we describe the use of a commercially available photoacoustic (PA) imaging system (Vevo LAZR, VisualSonics, Toronto) to image inducible gene expression in subcutaneous xenograft tumors using tyrosinase as a reporter gene. The photoacoustic imaging system generated light from a tunable laser (680 - 970 nm) which was delivered through fiber optic bundles integrated into a linear array transducer (LZ-250, fc = 21 MHz), mounted to a linear stepper motor for 3D imaging. Animals (n=3) having MCF-7 xenograft tumors on either flank transfected with (+TYR) or without (-TYR) doxycycline-regulated tyrosinase were imaged before and one week after the induction of tyrosinase expression. 3D images of the tumors were acquired using multiple wavelengths (680, 750, 800, 850, 900 and 950nm) and 2D images were acquired across the entire wavelength range of the laser to generate absorption curves for blood and melanin. To confirm the presence of melanin and distinguish it from the endogenous blood signal, one animal was exsanguinated and the tumor was imaged again. Approximately 1mm-thick slices of the tumors were taken and photographed for visual confirmation of the presence of melanin. Comparison of pre- and post-doxycycline images of +TYR tumors clearly showed enhanced contrast in the tumor which closely matched the absorption spectrum of melanin. This signal persisted upon exsanguination. -TYR tumors showed signal which corresponded with the spectra of oxy and deoxy hemoglobin and changed little over the course of the experiment. Visual inspection of the excised sliced tumors revealed pigmentation in the +TYR tumors which was absent in the -TYR tumors. Here we have shown the ability of the Vevo LAZR photoacoustic imaging system to visualize inducible reporter gene expression in vivo. This has implications for assessing genetically controlled cellular processes and their response to anti-cancer drugs but also for monitoring gene therapy for cancer treatment non-invasively. Photoacoustic (PA) imaging systems are spreading in the biomedical community, and the development of new PA contrast agents is an active area of research. However, PA contrast agents are usually characterized with spectrophotometry or uncalibrated PA imaging systems, leading to partial assessment of their PA efficiency. To enable quantitative PA spectroscopy of contrast agents in vitro with conventional PA imaging systems, we have developed an adapted calibration method. Contrast agents in solution are injected in a dedicated non-scattering tube phantom imaged at different optical wavelengths. The calibration method uses a reference solution of cupric sulfate to simultaneously correct for the spectral energy distribution of excitation light at the tube location and perform a conversion of the tube amplitude in the image from arbitrary to spectroscopic units. The method does not require any precise alignment and provides quantitative PA spectra, even with non-uniform illumination and ultrasound sensitivity. It was implemented on a conventional imaging setup based on a tunable laser operating between 680 nm and 980 nm and a 5 MHz clinical ultrasound array. We demonstrated robust calibrated PA spectroscopy with sample volumes as low as 15 μL of known chromophores and commonly used contrast agents. The validated method will be an essential and accessible tool for the development of new and efficient PA contrast agents by improving their quantitative characterization.

ObjectiveTo evaluate in vivo a fully integrated photoacoustic tomography imaging system based on Fabry–Pérot ultrasound sensing method applied on porcine abdominal organs. This approach could be used by surgeons during intraoperative clinical procedures.MethodsThe photoacoustic imaging system was fully integrated into a single structure, and the detection technology was based on a Fabry–Pérot interferometer. The detection probe connected to the imaging system was applied directly to the organs of a male “large white” Sus scrofa pig weighing 80 kg, either manually or using a stand, with or without a gel interface. All experiments were performed in compliance with EU Directive 2010/63/EU on animal experimentation (APAFiS #31507).ResultsAll intraperitoneal and retroperitoneal organs were evaluated using photoacoustic imaging. The evaluation of both hollow and solid organs was successfully conducted with consistent three-dimensional image quality. We demonstrate the system’s ability to image blood vessels with diameters ranging from several millimeters down to less than 100 µm. Macroscopic evaluation of the organs using photoacoustic tomography imaging did not reveal any damage or burns caused by the excitation laser.ConclusionTo our knowledge, this is the first reported imaging session of abdominal organs in an in vivo porcine model, performed using a photoacoustic tomography system with Fabry–Pérot interferometer detection. Photoacoustic (PA) imaging provides excellent optical contrast with decent penetration and high spatial resolution, making it attractive for a variety of neural applications. We evaluated optical contrast agents with high absorption in the near infrared (NIR) as potential enhancers for PA neuroimaging: optical dyes, gold nanorods (GNRs) and PEBBLEs loaded with indocyanine green. Two PA systems were developed to test these agents in excised neural tissue and in vivo mouse brain. Lobster nerves were stained with the agents for 30 minutes and placed in a hybrid nerve chamber capable of electrical stimulation and recording, optical spectroscopy and PA imaging. Contrast agents boosted the PA signal by at least 30 dB using NIR illumination from a tunable pulsed laser. Photobleaching may be a limiting factor for optical dyes-the PA signal decreased steadily with laser illumination. The second setup enabled in vivo transcranial imaging of the mouse brain. A custom clinical ultrasound scanner and a 10-MHz linear array provided near real-time images during and after an injection of 2 nM gold nanorods into the tail vein. The peak PA signal from the brain vasculature was enhanced by up to 2 dB at 710 nm. Temporal dynamics of the PA signal were also consistent with mixing of the GNRs in the blood. These studies provide a baseline for enhanced PA imaging in neural tissue. The smart contrast agents employed in this study can be further engineered for molecular targeting and controlled drug delivery with potential treatment for a myriad of neural disorders.

Understanding the tumor microenvironment is critical to characterizing how cancers operate and predicting their response to treatment. We describe a novel, high-resolution coregistered photoacoustic (PA) and pulse echo (PE) ultrasound system used to image the tumor microenvironment. Compared to traditional optical systems, the platform provides complementary contrast and important depth information. Three mice are implanted with a dorsal skin flap window chamber and injected with PC-3 prostate tumor cells transfected with green fluorescent protein. The ensuing tumor invasion is mapped during three weeks or more using simultaneous PA and PE imaging at 25 MHz, combined with optical and fluorescent techniques. Pulse echo imaging provides details of tumor structure and the surrounding environment with 100-μm(3) resolution. Tumor size increases dramatically with an average volumetric growth rate of 5.35 mm(3)/day, correlating well with 2-D fluorescent imaging (R = 0.97, p < 0.01). Photoacoustic imaging is able to track the underlying vascular network and identify hemorrhaging, while PA spectroscopy helps classify blood vessels according to their optical absorption spectrum, suggesting variation in blood oxygen saturation. Photoacoustic and PE imaging are safe, translational modalities that provide enhanced depth resolution and complementary contrast to track the tumor microenvironment, evaluate new cancer therapies, and develop molecular contrast agents in vivo.

In this system, PA signals originating from blood in the noninjured tissue layer located under the injured tissue layer are detected. The phantom study showed that thin light absorbers embedded in the tissue-mimicking scattering medium at depths of > 3 mm can be imaged with high contrast. The diagnostic experiments using rat burn models showed good agreements between the injury depths (zones of stasis) indicated by PA imaging and those by histological analysis. These results demonstrate the potential usefulness of the present system for clinical burn diagnosis.

We have developed a real-time (8 to 30 fps) photoacoustic (PA) imaging system with a linear-array transducer for burn depth assessment. In this system, PA signals originating from blood in the noninjured tissue layer located under the injured tissue layer are detected and imaged. A compact home-made high-repetition-rate (500 Hz) 532-nm fiber laser was incorporated as a light source. We used an alternating arrangement for the fibers and sensor elements in the probe, which improved the signal-to-noise ratio, reducing the required laser energy power for PA excitation. This arrangement also enabled a hand-held light-weight probe design. A phantom study showed that thin light absorbers embedded in the tissue-mimicking scattering medium at depths >3 mm can be imaged with high contrast. The maximum error for depth measurement was 140 μm. Diagnostic experiments were performed for rat burn models, including superficial dermal burn, deep dermal burn, and deep burn models. Injury depths (zones of stasis) indicated by PA imaging were compared with those estimated by histological analysis, showing discrepancies 200 μm. The system was also used to monitor the healing process of a deep dermal burn. The results demonstrate the potential usefulness of the present system for clinical burn diagnosis.

Introduction: There is an urgent clinical need to develop noninvasive biomarkers that early detect tumor responses to anti-angiogenic therapy. Treatment response in ovarian cancer patients is assessed with transvaginal ultrasound imaging of tumor size and CA125 screening. We hypothesized that combining photoacoustic (PA) imaging to non-invasively visualize tumor vascular architecture, with serum diagnostics using endothelial biomarkers, would yield a sensitive and readily translatable approach for monitoring response to anti-angiogenic therapy. Methods: Ovarian tumors were established in nude mice by orthotopic injection of OV2008 cells at day 0. At day 10 mice were randomized into groups of at least n = 10. Baseline PA imaging and submandibular blood draw were performed, then repeated 24h after dosing with a peptibody that prevents the interaction of secreted angiopoietin 1 (Ang1; agonist) and 2 (Ang2; antagonist) with the receptor tyrosine kinase Tie2, or vehicle, on days 13, 16 and 19. Mice were then sacrificed and tumors excised for histology. Results: PA imaging uses endogenous hemoglobin light absorption to generate contrast. At 797nm, we found a 1.5-fold increase in hemoglobin-weighted signal in vehicle mice by day 19, while treated mice remained close to the baseline (n=9; p=0.008). The oxyhemoglobin-weighted PA signal (837nm/797nm) was elevated by 20% in treated mice but decreased 15% in vehicle mice (n=9; p&amp;lt;0.01). This indicated treated mice had significantly lower tumor vessel density but improved function of residual vessels. To gain insight into the mechanism of drug action, we measured host serum angiopoietins over the treatment time course. Ang1 in vehicle mice increased by 1.5-fold (n=10; p&amp;lt;0.0001) and was significantly higher than treated mice by day 19 (n=10; p=0.03). Ang2 was identical between treatment groups and healthy control mice (n=10; p=0.65). Elevated Ang1 in the circulation of vehicle mice is consistent with excess Ang1 promoting vascular remodeling and plasticity in tumors, while normalization in treated mice allows vessel stabilization. Immunofluorescence (IF) confirmed this by indicating that CD31 positive vessel density was 30% lower in treated mice but their tumor vessel coverage of desmin-positive pericytes was 2.5-fold higher. Conclusions: We have shown for the first time that PA imaging and serum angiopoietin levels measure complementary parameters that indicate response to angiopoietin blockade in ovarian cancer. PA imaging noninvasively detected drug-induced vessel regression and normalization, giving a functional readout of response. This modality can be readily combined with transvaginal ultrasound. The drug action was mediated by normalization of mAng1 levels, indicated by serum measures and IF. Importantly, this work supports the combined use of in vitro and in vivo diagnostics to monitor response to anti-angiogenic therapy in clinical trials. Citation Format: Sarah E. This paper presents a combined ultrasound and photoacoustic (PA) imaging (PAI) system used to obtain high-quality, co-registered images of mouse-embryo anatomy and vasculature. High-frequency ultrasound (HFU, >20 MHz) is utilized to obtain high-resolution anatomical images of small animals while PAI provides high-contrast images of the vascular network. The imaging system is based on a 40 MHz, 5-element, 6 mm aperture annular-array transducer with a 800 μm diameter hole through its central element. The transducer was integrated in a cage-plate assembly allowing for a collimated laser beam to pass through the hole so that the optical and acoustic beams were collinear. The assembly was mounted on a two-axis, motorized stage to enable the simultaneous acquisition of co-registered HFU and PA volumetric data. Data were collected from all five elements in receive and a synthetic-focusing algorithm was applied in post-processing to beamform the data and increase the spatial resolution and depth-of-field (DOF) of the HFU and PA images. Phantom measurements showed that the system could achieve high-resolution images (down to 90 μm for HFU and 150 μm for PAI) and a large DOF of >8 mm. Volume renderings of a mouse embryo showed that the scanner allowed for visualizing morphologically precise anatomy of the entire embryo along with corresponding co-registered vasculature. Major head vessels, such as the superior sagittal sinus or rostral vein, were clearly identified as well as limb bud vasculature.
