Vascularization in the primate visual cortex during development.

Predicting the Stream of Consciousness from Activity in Human Visual Cortex

Low-intensity ultrasound stimulation of the retina has the ability to modulate neural activity in the primary visual cortex (V1), however, it is currently unclear how different intensities and durations of ultrasonic stimulation of the retina modulate neural activity in V1. In this paper, we recorded local field potential (LFP) signals in the V1 brain region of mice under different ultrasound intensities and different stimulation times. The amplitude of LFP corresponding to 1 s before ultrasound stimulation to 2 s after stimulation (-1-2 s) was analyzed, including the power and sample entropy of delta, theta, alpha beta, and low gamma frequency bands. The experimental results showed that, as the stimulation intensity increased, the peak value of the LFP in the visual cortex showed a linear upward trend; the power in the delta and theta frequency bands showed a linear upward trend, and the sample entropy showed a linear downward trend. With increases of stimulation duration, the peak value of the LFP in the visual cortex showed an upward trend, and the upward trend gradually weakened; the power in the delta frequency band showed an upward trend, the sample entropy showed a linear upward trend, and the sample entropy in the theta frequency band showed a downward trend. The results show that low-intensity ultrasonic stimulation of the retina has a significant modulatory effect on neural activity in the visual cortex. The study provides insights into the mechanisms by which ultrasonic stimulation regulates visual system function. Furthermore, it clarifies the patterns of parameter selection, facilitating the development of personalized multi-parameter modulation for the treatment of visual neural degeneration, retinal disorders and related research areas.

Positron emission tomography was used to measure cerebral activity and to evaluate regional interrelationships within visual cortices and their projections during rapid eye movement (REM) sleep in human subjects. REM sleep was associated with selective activation of extrastriate visual cortices, particularly within the ventral processing stream, and an unexpected attenuation of activity in the primary visual cortex; increases in regional cerebral blood flow in extrastriate areas were significantly correlated with decreases in the striate cortex. Extrastriate activity was also associated with concomitant activation of limbic and paralimbic regions, but with a marked reduction of activity in frontal association areas including lateral orbital and dorsolateral prefrontal cortices. This pattern suggests a model for brain mechanisms subserving REM sleep where visual association cortices and their paralimbic projections may operate as a closed system dissociated from the regions at either end of the visual hierarchy that mediate interactions with the external world.

Neural activity in human visual cortex is transformed by learning real world size

When and where are decisions made? In the visual system a saccade, which is a fast shift of gaze toward a target in the visual scene, is the behavioral outcome of a decision. Current neurophysiological data and reaction time models show that saccadic reaction times are determined by a build-up of activity in motor-related structures, such as the frontal eye fields. These structures depend on the sensory evidence of the stimulus. Here we use a delayed figure-ground detection task to show that late modulated activity in the visual cortex (V1) predicts saccadic reaction time. This predictive activity is part of the process of figure-ground segregation and is specific for the saccade target location. These observations indicate that sensory signals are directly involved in the decision of when and where to look.

The aim of this study was to examine the secondary visual cortex functional disorder in patients with glaucoma and large pituitary adenoma by functional magnetic resonance imaging, and to determine the correlation between visual field defect and primary visual cortex activation. Results showed that single eye stimulation resulted in bilateral visual cortex activation in patients with glaucoma or large pituitary adenoma. Compared with the normal control group, the extent and intensity of visual cortex activation was decreased after left and right eye stimulation, and functional magnetic resonance imaging revealed a correlation between visual field defects and visual cortex activation in patients with glaucoma and large pituitary adenoma. These functional magnetic resonance imaging data suggest that anterior optic pathway lesions can cause secondary functional disorder of the visual cortex, and that visual defects are correlated with visual cortex activation.

We recently obtained the brain of a rare lemuroid primate, Cheirogaleus medius. The brain was not perfused before death, but rather fixed by immersion shortly thereafter. In both flat-mounted and transversely sectioned tissue, we were able to clearly demonstrate periodic zones of high cytochrome oxidase (CO) activity in the primary visual cortex, resembling the so-called 'blobs' described in many other primate species. Our results contrast with a previous report indicating that blobs are absent in Cheirogaleus medius and provide support for the view that blobs are an evolutionary specialization of primate visual cortex that evolved only once, early in primate history. In other aspects of architectonic organization, area V1 of this Cheirogaleus individual closely resembles that of other strepsirhine primates, such as Galago. We were able to identify additional divisions of cortex in this individual, including the middle temporal visual area (MT), auditory cortex, and the primary somatosensory area (S1 or area 3b). These observations indicate that valuable neuroanatomical information can, in favorable cases, be obtained from rare mammalian species that die of natural causes in captivity or which must be euthanized, even though the animals have not been perfused.

Evidence from human neuroimaging and animal electrophysiological studies suggests that signals from different sensory modalities interact early in cortical processing, including in primary sensory cortices. The present study aimed to test whether functional near-infrared spectroscopy (fNIRS), an emerging, non-invasive neuroimaging technique, is capable of measuring such multisensory interactions. Specifically, we tested for a modulatory influence of sounds on activity in visual cortex, while varying the temporal synchrony between trains of transient auditory and visual events. Related fMRI studies have consistently reported enhanced activation in response to synchronous compared to asynchronous audiovisual stimulation. Unexpectedly, we found that synchronous sounds significantly reduced the fNIRS response from visual cortex, compared both to asynchronous sounds and to a visual-only baseline. It is possible that this suppressive effect of synchronous sounds reflects the use of an efficacious visual stimulus, chosen for consistency with previous fNIRS studies. Discrepant results may also be explained by differences between studies in how attention was deployed to the auditory and visual modalities. The presence and relative timing of sounds did not significantly affect performance in a simultaneously conducted behavioral task, although the data were suggestive of a positive relationship between the strength of the fNIRS response from visual cortex and the accuracy of visual target detection. Overall, the present findings indicate that fNIRS is capable of measuring multisensory cortical interactions. In multisensory research, fNIRS can offer complementary information to the more established neuroimaging modalities, and may prove advantageous for testing in naturalistic environments and with infant and clinical populations.

To test the hypothesis that calcitonin gene-related peptide (CGRP) modulates brain activity, we investigated the effect of intravenous CGRP on brain activity in response to a visual stimulus. In addition, we examined if possible alteration in brain activity was reversed by the anti-migraine drug sumatriptan. Eighteen healthy volunteers were randomly allocated to receive CGRP infusion (1.5 μg/min for 20 min) or placebo. In vivo activity in the visual cortex was recorded before, during and after infusion and after 6 mg subcutaneous sumatriptan by functional magnetic resonance imaging (3 T). 77% of the participants reported headache after CGRP. We found no changes in brain activity after CGRP (P = 0.12) or after placebo (P = 0.41). Sumatriptan did not affect brain activity after CGRP (P = 0.71) or after placebo (P = 0.98). Systemic CGRP or sumatriptan has no direct effects on the BOLD activity in visual cortex. This suggests that in healthy volunteers both CGRP and sumatriptan may exert their actions outside of the blood–brain barrier.Electronic supplementary materialThe online version of this article (doi:10.1007/s10194-011-0415-4) contains supplementary material, which is available to authorized users.

To visualize dreaming brain functions we studied hemodynamic changes in the visual cortex during the transition from non-rapid eye movement (NREM) to rapid eye movement (REM) sleep, using a 24-channel Near-Infrared Spectroscopy (NIRS) imaging method. Results were compared to the activation in visual cortex by visual stimulation during wakefulness. Subjects were four healthy males between 25 and 49 years of age. Five all-night polysomnographic and NIRS recordings were made. Increases in the oxygenated hemoglobin concentration in visual cortex were observed from nine of 14 REM periods. The activated areas were broader during REM sleep than during visual stimulation. These findings suggest that activation of visual cortex in REM sleep might represent dream-related brain activity.

Brain mitochondrial cytochrome oxidase and respiratory activities were compared after in vivo and in vitro exposure to cyanide. For the in vivo studies, mice were exposed to a non-lethal (4 mg kg-1) or lethal (20 mg kg-1) dose of KCN. From these mice, purified brain mitochondria were prepared and cytochrome oxidase and respiratory activities measured. Results of these experiments revealed greater inhibition of cytochrome oxidase activity following a lethal (20 mg kg-1) than a non-lethal (4 mg kg-1) KCN dose (57 and 45% inhibition, respectively). Respiration states 3 and 4 of brain mitochondria prepared from mice that received 4 mg kg-1 KCN were inhibited by 15 and 20%, respectively. In mice that received a lethal 20 mg kg-1 KCN dose, respiration states 3 and 4 were each inhibited by ca. 30% (P < 0.05). In vitro, mitochondrial cytochrome oxidase activity was inhibited in a concentration-dependent fashion at cyanide concentrations of 10(-6)-10(-2) M. A biphasic inhibition of ADP-stimulated (state 3) respiration was observed. Cyanide concentrations of 10(-6)-10(-4) M produced only a 25% inhibition of respiration state 3, whereas 10(-3) M produced 80% inhibition. Because this dramatic inhibition only occurred at cyanide concentrations that caused > 50% inhibition of mitochondrial cytochrome oxidase activity, these findings suggest that a large proportion of cytochrome oxidase activity may be functional reserve and that cyanide poisoning likely involves other mechanisms in addition to inhibition of cytochrome oxidase.

Platelet cytochrome oxidase and citrate synthase activities in APOE ε4 carrier and non-carrier Alzheimer's disease patients

Cocaine is one of the most widely used illicit drugs in the world, and as a result of incomplete removal by sewage treatment plants it is found in surface waters, where it represents a new potential risk for aquatic organisms. In this study we evaluated the influence of environmental concentrations of cocaine on the liver and the kidney of the European eel (Anguilla anguilla). The eels were exposed to 20 ng L−1 of cocaine for fifty days, after which, three and ten days after the interruption of cocaine exposure their livers and kidneys were compared to controls. The general morphology of the two organs was evaluated, as well as the following parameters: cytochrome oxidase (COX) and caspase-3 activities, as markers of oxidative metabolism and apoptosis activation, respectively; glucose-regulated protein (GRP)78 levels, as a marker of endoplasmic reticulum (ER)-stress; blood glucose level, as stress marker; serum levels of alanine aminotransferase (ALT), as a marker of liver injury and serum levels of C-reactive protein (CRP), as a marker of the inflammatory process. The liver showed morphologic alterations such as necrotic areas, karyolysis and pyknotic nuclei, while the kidneys had dilated glomeruli and the renal tubules showed pyknotic nuclei and karyolysis. In the kidney, the alterations persisted after the interruption of cocaine exposure. In the liver, COX and caspase-3 activities increased (COX: P = 0.01; caspase-3: P = 0.032); ten days after the interruption of cocaine exposure, COX activity returned to control levels (P = 0.06) whereas caspase-3 activity decreased further (P = 0.012); GRP78 expression increased only in post-exposure recovery specimens (three days: P = 0.007 and ten days: P = 0.008 after the interruption of cocaine exposure, respectively). In the kidney, COX and caspase-3 activities increased (COX: P = 0.02; caspase-3: P = 0.019); after the interruption of cocaine exposure, COX activity remained high (three days: P = 0.02 and ten days: P = 0.029 after the interruption of cocaine exposure, respectively) whereas caspase-3 activity returned to control values (three days: P = 0.69 and ten days: P = 0.67 after the interruption of cocaine exposure, respectively). Blood glucose and serum ALT and CRP levels increased (blood glucose: P = 0.01; ALT: P = 0.001; CRP: 0.015) and remained high also ten days after the interruption of cocaine exposure (blood glucose: P = 0.009; ALT: P = 0.0031; CRP: 0.036). These results suggest that environmental cocaine concentrations adversely affected liver and kidney of this species.

Correlation between cytochrome oxidase staining and the uptake and laminar distribution of tritiated aspartate, glutamate, γ-aminobutyrate and glycine in the striate cortex of the squirrel monkey

Neuroscience Hearing sounds helps our visual system to predict incoming information and may give us a survival advantage. Vetter et al. blindfolded people and scanned their brains while they listened to birds singing, traffic noise, or people talking. Using sophisticated algorithms, the researchers were able to identify the category of sounds just by examining the pattern of activity in the primary visual cortex, a brain area previously believed to process nothing but input from the eyes. And when the people imagined the specific sound categories in the complete absence of sight and sound, their primary visual cortices also showed activity.
