W 1976 r. w Instytucie Genetyki, Hodowli i Biotechnologii Roślin Akademii Rolniczej w Lublinie (obecnie Uniwersytetu Przyrodniczego) rozpoczęto prace mające na celu gromadzenie i ocenę kolekcji pszenicy twardej. Ocena obiektów jest prowadzona w 3-letnim cyklu doświadczeń polowych, które po wieloletniej waloryzacji przekazywane są do klimatyzowanej przechowalni IHAR w Radzikowie w celu zabezpieczenia ich żywotności. Badania dotyczą testowania światowych genotypów w warunkach glebowo-klimatycznych naszego kraju. Dotychczas zgromadzono 2072 obiekty jarej pszenicy twardej. Zgromadzone materiały kolekcyjne pochodzą z ponad 40 krajów. Dość znaczną grupę stanowią formy pochodzące z Meksyku, Egiptu, Włoch oraz z krajów byłego Związku Radzieckiego. Zgromadzone genotypy reprezentują ok. 50 różnych odmian botanicznych, największą grupę obiektów stanowią takie odmiany botaniczne, jak: var. leucurum, var. leucomelan oraz var. hordeiforme, które łącznie stanowią ponad 50% zgromadzonej kolekcji pszenicy twardej i są przedmiotem opracowania. Przedstawione wyniki dotyczą średnich wieloletnich ważniejszych cech użytkowych, w tym cech plonotwórczych kłosa oraz oceny polowej. Z badań wynika, że zgromadzone genotypy charakteryzują się dużą zmiennością pod względem omawianych cech. Pomiędzy poszczególnymi cechami plonotwórczymi zachodzą różnego rodzaju korelacje. Genotypy charakteryzujące się dużą masą ziarn z kłosa mają na ogół dużą masę 1000 ziarn, co potwierdzają wysokie współczynniki korelacji, a genotypy o dużej masie 1000 ziarn wykazują mniejszą zawartość białka ogólnego w ziarnie, gdyż korelacja między tymi cechami jest ujemna.

The 3-year experiment was concerned with the response of spring forms of common wheat (Triticum aestivum L. subsp. aestivum), durum wheat (Triticum durum Desf.) and spelt wheat (Triticum aestivum subsp. spelta L. em. Thell.) to the foliar application of a plant growth stimulant (extract from marine algae Ecklonia maxima), with the commercial name of Kelpak SL (GS), as compared to control treatment (C). The following parameters were analysed: yield of grain, yield components (number of ears, weight of 1000 kernels, number and weight of kernels per ear) and physical indicators of grain quality (test weight, uniformity and vitreosity of grain). The study showed that the level of yielding and the yield components were related primarily with the wheat genotype, but they depended also on the agro-climatic conditions and on the algae extract and control experimental treatments. The application of algae extract, compared to the control, caused a significant increase in the yields of the spring wheat species under study, on average by 7.0%. Canopy spraying with algae extract had a favourable effect on the number of ears, on he number and weight of kernels per ear, but it had no effect on the weight of 1000 kernels. The grain quality of durum wheat, spelt wheat and common wheat was affected more strongly by the weather conditions in the successive years of the study and by the genotype than by the foliar application of algae extract. The spelt genotypes were characterised by lower yields and lower grain quality than common wheat and the durum wheat genotypes.

Since the publication of the full sequence of the Arabidopsis thaliana genome in 2000 [AGI Initiative 2000], a period of dynamic genome exploration began. In the last decade, with the revolution in the next generation sequencing, the number of scientific reports based on sequence analysis has increased exponentially. New, fast, high throughput and relatively inexpensive nucleic acid sequencing technologies have become available and widespread, opening up the possibility of making extensive use of molecular tools in science and breeding practice. These new methods include whole genome sequencing and various methods of reduced representation sequencing. The multitude of available methods, varying in terms of availability and costs, generating different types of result data, dedicated to different research and application purposes, may make it difficult to choose the best variant [Poland et al. 2012]. The aim of this paper is to familiarize the reader with the possibilities and application of selected genotyping techniques with the use of sequencing.

The objective of the study was to analyze the changes that took place on the Polish cereal market in the years 1965–2015 in the structure of sowing area, yields and harvests. The analysis included such cereals as wheat, rye, triticale, barley, oat, maize, buckwheat and millet, as well as cereal mixtures. Over the fifteen-year period under analysis fundamental changes were observed in the structure of the area of sowings, harvests, and yields of the cereals. The share of cereals with higher productivity (wheat, triticale, barley, maize) was gradually increasing and the area of rye and oat decreased. In the final 15 years the area of cultivation of cereals decreased, while the harvests and yields increased. The increase was especially notable in the share of cereals with a high production potential (wheat, barley, maize, triticale). In Poland, wheat is the cereal with the greatest importance for agriculture – in the analysed period it produced the highest harvests and yields as compared to other cereals. After 1990, the factor that contributed to the increase in the sowing area and harvests was the introduction of a new cereal species – triticale – in a wide agricultural practice. The variation in the harvests and yields was caused primarily by changes in sowings area, weather conditions, and mineral fertilization. The increase of the yields of cereals was caused by advances in biology (breeding of new cultivars), change in the configuration of cereals cultivation, and improved effectiveness of application of fertilizers and plant protection agents. The highest increasing trend was characteristic of the yields of wheat, and the strongest variation in the yields of cereals in Poland in the years 1965–2015 was characteristic of the yields of maize (30.3%).

Proekologiczne metody ograniczania występowania agrofagów w agrofitocenozach, polegające na wprowadzaniu do uprawy międzyplonów, wysiewie odmian konkurencyjnych względem chwastów oraz stosowaniu integrowanej ochrony roślin, sprzyjają zachowaniu bioróżnorodności obszarów wiejskich. Wyniki badań wskazują, iż wymienione zabiegi agrotechniczne mogą skutecznie ograniczać występowanie chwastów w agrocenozach, nie zmniejszając jednocześnie ich różnorodności biologicznej. Redukują występowanie kosmopolitycznych taksonów dominujących, zmniejszając ich liczebność poniżej ekonomicznego progu szkodliwości. Skuteczność tych zabiegów zależy jednak od wielu czynników siedliskowych i agrotechnicznych, takich jak typ gleby, warunki pogodowe, gatunek rośliny uprawnej, płodozmian, rodzaj międzyplonu i sposób jego zagospodarowania. Technologia produkcji uwzględniająca wymienione, przyjazne dla środowiska metody ograniczania występowania agrofagów, ma na celu symbiozę celów produkcyjnych i ekologicznych.

Pszenica należy do najważniejszych roślin zbożowych i jest powszechnie wykorzystywana w celach konsumpcyjnych. Obecnie prowadzi się silną selekcję odmian pod względem poprawy jej cech jakościowych. Jedną z ważniejszych cech pszenicy, decydującą o jej późniejszym wykorzystaniu, jest zawartość białek, które determinują właściwości lepkosprężyste ciasta pszennego. Właściwości reologiczne glutenu warunkowane są przez ilość i kompozycję białek zapasowych (glutenin i gliadyn) wchodzących w jego skład, przy czym białka gluteninowe wykazują znacząco większy wpływ na korzystne właściwości reologiczne. Poznanie ich składu podjednostkowego w ziarnie pszenicy, szczególnie podjednostek glutenin wysokocząsteczkowych (HMW – GS), umożliwia wybór genotypów o pożądanych właściwościach reologicznych na wczesnym etapie selekcji, co pozwala na szybsze i dokładniejsze osiągnięcie założonych przez hodowców celów. Rozwój technologii pozwolił na wykorzystanie nowych metod identyfikacji składu podjednostkowego glutenin. W tym celu obecnie stosuje się metody SDS-PAGE, RP-HPLC, HPCE, MALDI-TOF-MS oraz PCR.
