Identification of Intronless Genes and the Development of KASP Markers for Salt Responses in Vicia faba L.
Background/Objectives: Salinity stress limits agricultural production and threatens global food security. Faba bean (Vicia faba L.) is an important legume crop, and identifying salt-stress-responsive genes may support an improvement in salt response. This study aimed to identify intronless genes in faba bean, screen candidate genes associated with salt-stress responses, and develop a KASP marker for salt-response evaluation. Methods: Intronless genes were identified from the faba bean reference genome. Transcriptome analysis was conducted in roots and leaves of two cultivars, Sucan 4 and Yundou 1183, under 150 mM NaCl treatment and control conditions. Candidate genes were examined by expression analysis, functional annotation, PPI prediction, and a luciferase complementation assay. A KASP marker was developed from an SNP within the VfERF1A locus and tested in 97 accessions. Results: A total of 7581 intronless genes were identified, accounting for 20.69% of annotated genes. Fifteen intronless genes were significantly differentially expressed in both roots and leaves of the two cultivars under salt treatment. Functional annotation suggested that VfERF1A and VfHSP17.8 may be involved in salt-stress responses. PPI prediction and the LUC assay provided preliminary support for a possible association of VfERF1A with VfEIN2. The VfERF1A-based KASP marker showed clear genotype clustering, and the two homozygous classes differed significantly in QYmax, relative shoot fresh weight, and relative plant height under salt treatment (p < 0.05). The preliminary predictive accuracy for QYmax was 86.36%. Conclusions: These results provide a genome-wide resource of intronless genes in faba bean, identify candidate genes associated with salt-stress responses, and describe a preliminary KASP marker associated with salt-response traits. Further validation in independent populations, under diverse environmental conditions, and with additional functional evidence is still required.

To explore the characteristic volatile compounds of 'Hujing Milu' peaches from different growing regions, headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry (HS-SPME-GC-MS) and headspace gas chromatography-ion mobility spectrometry (HS-GC-IMS) were employed to analyze volatile components in samples from six production areas. A total of 73 and 56 volatile compounds were identified by HS-SPME-GC-MS and HS-GC-IMS, respectively. Quantitative analysis revealed that esters, aldehydes, and alcohols were the main contributors to the aroma profile, accounting for over 70% of the total relative content. Combined with chemometric analysis (VIP > 1 and OAV/ROAV > 1), 17 potential biomarkers were identified that can distinguish 'Hujing Milu' peaches from different regions, including ethyl acetate, hexanol, (E)-2-nonenal, and dihydro-β-ionone. Moreover, soil properties of these regions and their correlation with volatile compounds were analyzed to elucidate the formation mechanisms of characteristic aromas. The results showed that ethyl acetate exhibited a significant positive correlation with soil pH (r = 0.530, p < 0.05), whereas dihydro-β-ionone showed a significant positive correlation with soil organic matter (r = 0.587, p < 0.05) and available potassium (r = 0.830, p < 0.05). This study identified characteristic volatile compounds of 'Hujing Milu' peaches from different regions, providing a reliable technical basis for origin traceability and the enhancement of aroma quality in 'Hujing Milu' peaches.

With the escalating demand for forest-derived ecological products, quantifying forest ecological product production capacity (EPC) has become essential for precise ecological governance. Addressing the methodological gaps and complexity in current assessments, this study develops a transferable Forest EPC theoretical framework integrated across four dimensions: environmental background, vegetation status, human pressure, and human investment. Using Henan Province as a case study, we established a multi-criteria evaluation model to characterize the spatial drivers and supply potential of Forest EPC. Our findings reveal that the provincial forest EPC stands at a moderate level (0.341). The spatial distribution is highly heterogeneous: the “medium” EPC grade dominates the landscape (36.69%), whereas “high-level” areas are critically scarce (3.76%). Notably, forest EPC exhibits a strong spatial gradient, with high-performance clusters in the southern and western highlands contrasting with lower values in the northern plains. The identification of significant spatial autocorrelation (Global Moran’s I = 0.71) highlights the necessity of regional collaborative management. This study provides a methodological reference that is adaptable to diverse regional contexts through the recalibration of local indicators and weights, offering a scientific benchmark for optimizing the spatial layout of ecological product supply.

Selenium nanoparticles attenuate Klebsiella pneumoniae-induced mastitis via the suppression of ferroptosis and antioxidant restoration in goats.
Klebsiella pneumoniae-induced mastitis poses a severe threat to livestock health with limited therapeutic options. Selenium nanoparticles (SeNP) offer promising biocompatibility and antioxidant potential, but their efficacy and mechanism against bacterial mastitis remain unexplored. This study demonstrated that the oral administration of SeNP (20 μg/kg BW) attenuated K. pneumoniae-induced mastitis in goats. The treatment reduced histopathological damage and neutrophil infiltration. Furthermore, it restored anti-inflammatory IL-10 mRNA expression and suppressed proinflammatory IL-1β and IL-6 mRNA levels. Ultrasonography and Doppler analysis confirmed that mammary hemodynamics and echotextural abnormalities were improved. The SeNP supplementation contributed to increased concentrations of selenium in the serum, enhanced the activity of mammary glutathione, activated the peroxiredoxin 6 (PRDX6)/selenophosphate synthetase 2 (SEPHS2) axis, and upregulated glutathione peroxidase 4 (GPX4) expression. Immunofluorescence revealed that SeNP strengthened the PRDX6-SEPHS2 interaction to improve the activity of GPX4. Crucially, SeNP suppressed ferroptosis by inhibiting nuclear receptor coactivator 4 (NCOA4)-mediated ferritinophagy, reducing iron overload and lipid peroxidation, and attenuating NCOA4-ferritin heavy chain 1 colocalization. The findings established that SeNP concurrently target inflammatory, antioxidant, hemodynamic, and ferroptotic pathways to prevent bacterial mastitis, suggesting a novel intervention for the prevention and control of mastitis.
