POS0211 FIBROMYALGIA FOLLOW-UP BY FECAL AND PLASMA BIOMARKERS: A LOW-INTERVENTIONAL CLINICAL TRIAL
Study of the Risk Associated with Fish Ingestion in the Volta Grande of the Xingu River - Brazilian Amazon Region
Mining activity in the Amazon region has brought, over the years, environmental impacts and impacts on the health of traditional peoples such as indigenous and riverside peoples. The implementation of large projects in the region, such as hydroelectric plants, has changed the conditions of water bodies, adding potentially toxic metals to the environment that can be harmful to flora, fauna, and human beings. The objective of this research was to study the risk associated with local populations of ingestion of fish species captured in the Volta Grande region of the Xingu River, located in the Brazilian Amazon region. Sixty specimens of pacu (Piaractus mesopotamicus), pescada (Plagioscion spp.), piranha (Serrasalmus spp.), and tucunaré (Cichla spp.) fish were collected in two different regions of Volta Grande (middle and lower portions). For the determination of the elements Al, Mn, and Zn, an optical emission spectrometer with inductively coupled plasma was used, and for the Hg analysis, direct analysis with gold amalgamation was used. Statistical comparison tests and the use of indices were applied to assess the associated health risk, such as the Target Hazard Quotient (THQ) and Total Target Hazard Quotient (TTHQ) indicators and the risk of consuming food contaminated by Potentially Toxic Metals. The results showed that the elements Al, Mn, and Zn are the most susceptible to bioconcentration in fish, with the most affected species being Piaractus mesopotamicus and Serrasalmus spp. above safety limits.

Elastolysis and ineffective elastogenesis favor the accumulation of tropoelastin, rather than cross-linked elastin, in atherosclerotic plaques. We developed gadolinium-labeled tropoelastin-specific magnetic resonance contrast agents (Gd-TESMAs) for tropoelastin imaging in animal models. Two peptides, VVGSPSAQDEASPLS and YPDHVQYTHY were selected to target tropoelastin. In vitro binding, relaxivity, and biodistribution experiments enabled characterization of the probes and selecting the best candidate for in vivo MRI. MRI was performed in atherosclerotic apolipoprotein E-deficient (ApoE-/-) mice and New Zealand white rabbits with stable and rupture-prone plaques using Gd-TESMA. Additionally, human carotid endarterectomy specimens were imaged ex vivo. The VVGSPSAQDEASPLS-based probe discriminated between tropoelastin and cross-linked elastin (64±7% vs 1±2%, P=0.001), had high in vitro relaxivity in solution (r1-free=11.7±0.6mM-1s-1, r1-bound to tropoelastin = 44±1mM-1s-1) and favorable pharmacokinetics. In vivo mice vascular enhancement (4wks=0.13±0.007mm2, 8wks=0.22±0.01mm2, 12wks=0.33±0.01mm2, P<0.001) and R1 relaxation rate (4wks=0.90±0.01 s-1, 8wks=1.40±0.03 s-1, 12wks=1.87±0.04s-1, P<0.001) increased with atherosclerosis progression after Gd-TESMA injection. Conversely, statin-treated (0.13±0.01mm2, R1 =1.37±0.03s-1) and control (0.10±0.005mm2, R1 =0.87±0.05s-1) mice showed less enhancement. Rupture-prone rabbit plaques had higher R1 relaxation rate compared with stale plaques (R1=2.26±0.1s-1vs R1=1.43±0.02s-1, P=0.001), after administration of Gd-TESMA that allowed detection of rupture-prone plaques with high sensitivity (84.4%) and specificity (92.3%). Increased vascular R1 relaxation rate was observed in carotid endarterectomy plaques after soaking (R1pre= 1.1±0.26 s-1 vs R1post= 3.0±0.1s-1, P=0.01). Ex vivo analyses confirmed the MRI findings and showed uptake of the contrast agent to be specific for tropoelastin. MRI of tropoelastin provides a novel biomarker for atherosclerotic plaque progression and instability.

Influence of Additional Active Ingredients on the Effectiveness of Non‐Alcoholic Chlorhexidine Mouthwashes: A Randomized Controlled Trial
Non-alcoholic chlorhexidine mouthwashes are equally effective and have fewer potential risks than hydroalcoholic solutions. Nowadays, other active ingredients are added to these mouthwashes in an attempt to improve their effectiveness and reduce side effects. Following an experimental gingivitis model, this study examined three non-alcoholic commercial mouthwashes having 0.12% chlorhexidine digluconate (CHX) in common. Using a double masked, cross-over design, 30 subjects underwent three consecutive experimental phases with three mouthwashes: CHX, CHX + 0.05% sodium fluoride (CHXNaF), and CHX + 0.05% cetylpyridinium chloride (CHX-CPC). In each one of these 21-day phases, the subjects discontinued all oral hygiene measures and were treated exclusively with the experimental mouthwash randomly assigned (an oral rinse twice a day). Each experimental phase was preceded by a 14-day washout period. Levels of gingivitis, dental plaque, supragingival calculus, and dental staining were assessed at baseline and end (day 21) of experimental phases. The evolution of gingival and dental staining indices did not show statistically significant differences between the treatments. Differences were noticed in the plaque index (P = 0.0002), with CHX-NaF the treatment with the greatest increase. Differences were also observed in the supragingival calculus index (P = 0.0136), with CHX-CPC showing a smaller increase. Tongue staining was more frequent with CHX-CPC (P = 0.0141). In non-alcoholic 0.12% chlorhexidine mouthwashes, the addition of other active ingredients does not produce beneficial effects, but may even reduce the antiplaque effectiveness or increase tongue staining.
