Epidemiology and Video Analysis of Hamstring Injuries in the National Basketball Association
ABSTRACT Context: Hamstring injuries are common in the National Basketball Association (NBA), however there is a paucity of epidemiological data regarding these injuries. Objective: The primary aims of this study were to characterize the epidemiology and performance outcomes of hamstring injuries in the NBA over four seasons. Secondary aims were to analyze the mechanism of hamstring injury through video analysis. Design: Descriptive epidemiological study Setting: Publicly available injury reports, player statistics, and game footage from the NBA over four seasons (2019–2022). Patients or Other Participants: We included a total of 181 injuries sustained by NBA players for analysis. We analyzed available video film for 45 players to determine the mechanism of injury. Main Outcome Measure(s): We recorded player demographics and performance data. We used Game Score, which uses box score statistics such as points, rebounds, assists, steals and blocks to measure player performance. We compared performance outcomes by age, BMI, game score, and position using paired t-tests. Results: By position, guards experienced the highest proportion of injuries (65.7%). Players injured their left hamstring most frequently (59.6%; p=0.044). Players missed an average of 5.5 games after injury. Within 3 months of returning to play, 16.0% reinjured their ipsilateral hamstring and 5.0% injured their contralateral hamstring. Player performance declined after injury in the short and long-term by 1.1 and -0.8 game score units, respectively. Video analysis revealed most injuries were noncontact (62.2%) and occurred as a player initiated a plant/jump movement with the injured extremity (41.9%). Conclusions: High-performing players have higher rates of re-injury and a more significant decline in performance compared with low-performing players. Most injuries occurred on offense while the injured player was jumping or landing. Due to the frequency of hamstring injuries, awareness of injury mechanisms and characteristics of injury can be useful for risk mitigation purposes.

Return to Basketball After Primary Hip Arthroscopy for Femoroacetabular Impingement: Minimum 5-Year Follow-up
Background:Femoroacetabular impingement (FAI) is a common injury experienced by basketball players. Surgical management with arthroscopic labral repair and femoral osteochondroplasty can lead to successful outcomes for athletes whose nonoperative management fails. Recent studies have demonstrated that athletes undergoing hip arthroscopy in the setting of FAI had favorable return-to-sport (RTS) and sport continuation outcomes.Purpose:To evaluate patient-reported outcome measures (PROMs) and RTS rates at minimum 5-year follow-up in basketball players after hip arthroscopy for FAI.Study Design:Case series; Level of evidence, 4.Methods:All active basketball players at the recreational, high school, and collegiate levels who underwent primary hip arthroscopy with labral repair for FAI between August 2009 and December 2019 were identified. Postoperative RTS, revision rates, and surgery satisfaction at final follow-up were evaluated. PROMs were also analyzed as follows: visual analog scale, Tegner Activity Scale, modified Harris Hip Score, Hip Outcome Score–Activities of Daily Living, and Hip Outcome Score–Sport.Results:Fifty-three patients (64 hips) were included with a mean ± SD age of 21.2 ± 6.0 years. Most athletes played recreational or high school basketball (85%), while 15% played college basketball. Forty-three patients attempted to return to basketball. Of these patients, 35 (81%) returned to basketball after surgery, with 32 (91%) returning to the same level of play. Patients who returned to basketball had significantly improved outcomes on the modified Harris Hip Score (P = .006), Hip Outcome Score–Activities of Daily Living (P = .007), Hip Outcome Score–Sport (P = .004), Tegner Activity Scale (P = .010), and visual analog scale with use (P = .041) as compared with those who did not return. High school athletes had decreased odds of RTS when compared with recreational athletes (P = .01). There was significant improvement in all PROMs from baseline to minimum 5-year follow-up (P < .05).Conclusion:At minimum 5-year follow-up, basketball players demonstrated durable improvements in PROMs and an RTS rate of 81% after hip arthroscopy for FAI. The majority of athletes returned to the same level of competition. There were significantly improved PROMs in patients who returned to sport versus those who did not. High school athletes had decreased odds of RTS when compared with recreational athletes. However, body mass index, sex, age, alpha angle, and basketball position did not significantly correlate with RTS outcomes.

Abstract B035: CCT as a Driver of Prostate Cancer Invasion and Resistance: A New Therapeutic Target
Abstract Background: Prostate cancer (PCa) is the second leading cause of cancer-related death in men worldwide, with over 1.4 million new cases annually. Standard treatment for advanced disease includes androgen deprivation therapy (ADT), yet most patients progress to metastatic castration-resistant PCa (CRPC), where the 5-year survival rate falls below 30%. Current therapies targeting androgen receptor (AR) signaling or chemotherapy offer limited benefit, underscoring the need for novel strategies. Emerging data from castration-sensitive PCa (CSPC) and CRPC (unpublished) suggest that genomic instability increases during progression to CRPC, driving tumor heterogeneity and therapy resistance. The mechanisms underlying these changes remain poorly defined. We investigated the role of Chaperonin-Containing TCP-1 (CCT/TRiC), a protein-folding chaperone, in this process. Genomic instability during oncogenesis increases the production of misfolded proteins, potentially making CCT essential for PCa survival and adaptation under therapeutic stress. Methods: CCT is an eight-subunit complex (CCT1–8). Prior work identified the CCT2 subunit as critical for chaperonin oncogenic activity. Using PCa cell lines (LNCaP, PC3, 22Rv1), we depleted CCT2 via inducible small hairpin RNA (shRNA) and expressed FLAG-tagged CCT2 via lentiviral transduction. To therapeutically inhibit CCT, we developed an amphipathic peptide that targets the ATP-binding and oligomerization sites of CCT subunits and formulated a cell-permeable micellar version, Zwitterion-TRiC Off Peptide (Z-TOP). Functional assays assessed viability, proliferation, migration, and spheroid growth. Extracellular vesicles (EVs) released from PCa were isolated and characterized by nanoparticle tracking analysis (NTA) and flow cytometry. Transcriptomic profiling (RNA-seq) and pathway enrichment analysis were performed following CCT2 depletion or Z-TOP treatment. Results: CCT2 depletion or overexpression significantly altered PCa cell viability, growth, and migration. Z-TOP treatment produced similar effects, reducing viability, proliferation, migration, and spheroid size. Transcriptomic analysis revealed overlapping and distinct gene expression changes to cell cycle regulation, cancer signaling, and lipid pathways. EV studies indicated that CCT contributes to EV biogenesis; CCT2 mRNA and protein were detected in EV cargo. Both CCT2 depletion and Z-TOP treatment disrupted EV release and composition, potentially impairing metastatic signaling. Conclusions: CCT regulates PCa cell behavior and intercellular communication locally and via EVs, promoting hormone resistance and metastasis. These findings position CCT as a promising therapeutic target in advanced PCa. Z-TOP demonstrates potential for inhibiting metastasis and improving outcomes in CRPC. Future studies will evaluate Z-TOP in vivo and explore combination strategies with AR-targeted agents. Microsoft Co-Pilot was used to edit the text. Citation Format: Annette R.
