ANALYSIS OF STRENGTHENING THE CHARACTER OF ENTREPRENEURSHIP BASED ON LOCAL WISDOM AT SMA 1 KUDUS
This study aims to analyze and describe in detail the strengthening of the entrepreneurial character based on local wisdom that has been carried out by one of the schools in Kudus Regency, namely SMA 1 Kudus. This type of research is descriptive using qualitative methods. Data collection methods by conducting interviews, observations, and documentation. The data analysis used is an interactive data analysis technique by Miles, Huberman, and Saldana (2014) which consists of data condensation, data presentation, and conclusion drawing or verification. The results of this study show that planning to strengthen the entrepreneurial character based on local wisdom at SMA 1 Kudus is carried out in 3 ways, namely: through Craft and Entrepreneurship learning, through the P5 theme of entrepreneurship, and through the Intra-School Student Organization (OSIS). The implementation is carried out by optimizing the learning process, P5 activities, and student council to prioritize the processing of local natural products from Kudus Regency based on developing local values. Meanwhile, the evaluation of the activity found obstacles, one of which was in fulfilling financial needs during the activity. The conclusion of this research is that SMA 1 Kudus has succeeded in strengthening the entrepreneurial character of students, especially based on local wisdom
Read more“hDOS”: An automated hybrid diffuse optical device for real-time non-invasive tissue monitoring—precision andin vivovalidation
Abstract SignificanceA new platform/device is presented that advances hybrid diffuse optical monitors closed to clinical practice, bridging the gap between research-grade optical systems and practical bedside applications. Traditional devices often lack automation, multi-parameter functionality, and operator independence, hence, limiting their usability in demanding clinical environments. By offering automation, user-friendly operation, and overcoming the typical limitations of continuous-wave near-infrared spectroscopy, the hybrid diffuse optical platform (hDOS) provides a more accurate and reliable assessment of both oxygenation and perfusion. This innovation is particularly valuable for monitoring critically ill patients, where precise real-time measurements can directly influence patient management and outcomes.AimTo design, validate, and characterize the platform hDOS that integrates time-domain near-infrared spectroscopy, diffuse correlation spectroscopy, and a pulse oximeter with an automated vascular occlusion test (VOT). The platform aims to support continuous monitoring and the assessment of peripheral microvascular, and metabolic functions in both clinical and field settings.ApproachThe validation strategy for the hDOS device follows a comprehensive approach that goes beyond conventional optical performance assessments. Rather than solely verifying fundamental system parameters, the evaluation comprises of real-world usability, operator and patient safety, and clinical implementation. The device’s precision and usability were rigorously testedin vivothrough test-retest measurements and comparisons with a commercially available device (INVOS 5100C). This was subsequently followed by a seven-month clinical evaluation at Parc Taulí Hospital Universitari.ResultsThe device underwent extensive validation, accumulating over 200 hours of usage across approximately 150 measurement sessions. The hDOS device exhibited two-fold lower inter-subject and intra-subject variability in baseline tissue oxygen saturation compared to the INVOS 5100C. Furthermore, during a a vascular occlusion challenge, statistically significant differences were observed between the two systems across all extracted parameters. Finally, as a proof of concept, hDOS successfully detected differences in the microvasculature between a general mixed ICU patient cohort (n= 100) and a healthy control group (n= 37).ConclusionsOverall, hDOS device has performed well in both bench-top and realistic clinical applications on patientsin vivo. hDOS device provides a unique combination of parameters, available for the first time in a fully automated, self-contained platform.

Objective: To evaluate the impact of congenital scoliosis on spinal curvature severity and quality of life in patients treated at a single center in Pakistan, with a focus on demographic distribution, Cobb angle, and quality of life outcomes. Methodology: A retrospective cohort study was conducted from June 2013 to June 2023 at the Orthopedics and Spine Department, Hayatabad Medical Complex, Peshawar. The study included 100 patients aged 1 to 30 years diagnosed with congenital scoliosis. Data on demographics, Cobb angle measurements, and quality of life (using the SRS-22 questionnaire) were collected. Statistical analyses, including t-tests and chi-square tests, were performed to examine associations between gender, Cobb angle, and quality of life, with significance set at p < 0.05. Results: The mean age of participants was 15.2 years (SD = 8.4), with an even gender distribution (52% male). The average Cobb angle was 49.7 degrees (SD = 18.6), indicating moderate to severe scoliosis, with no significant difference between genders (mean Cobb angle: males = 51.2°, females = 48.1°; p = 0.26). Quality of life scores averaged 3.7 (SD = 0.9), and a moderate negative correlation was found between Cobb angle and quality of life score (r = -0.35), suggesting that more severe curvature is associated with lower quality of life. A chi-square test also indicated no significant association between gender and scoliosis severity (p = 0.99). Conclusion: This study emphasizes the impact of congenital scoliosis on quality of life, with severity of spinal curvature correlating with reduced well-being. The findings highlight the need for early diagnosis and targeted intervention to optimize outcomes. This foundational data contributes to the understanding and management of congenital scoliosis in similar healthcare settings.

Read morePharmacokinetics and Safety of Firsocostat, an Acetyl-Coenzyme A Carboxylase Inhibitor, in Participants with Mild, Moderate, and Severe Hepatic Impairment.
Firsocostat is an oral, liver-targeted inhibitor of acetyl-coenzyme A carboxylase in development for the treatment of metabolic dysfunction-associated steatohepatitis. Hepatic organic anion transporting polypeptides play a significant role in the disposition of firsocostat with minimal contributions from uridine diphospho-glucuronosyltransferase and cytochrome P450 3A enzymes. This phase 1 study evaluated the pharmacokinetics and safety of firsocostat in participants with mild, moderate, or severe hepatic impairment. Participants with stable mild, moderate, or severe hepatic impairment (Child-Pugh A, B, or C, respectively [n = 10 per cohort]) and healthy matched controls with normal hepatic function (n = 10 per cohort) received a single oral dose of firsocostat (20 mg for mild and moderate hepatic impairment; 5 mg for severe hepatic impairment) with intensive pharmacokinetic sampling over 96 h. Safety was monitored throughout the study. Firsocostat plasma exposure (AUCinf) was 83%, 8.7-fold, and 30-fold higher in participants with mild, moderate, and severe hepatic impairment, respectively, relative to matched controls. Firsocostat was generally well tolerated, and all reported adverse events were mild in nature. Dose adjustment is not necessary for the administration of firsocostat in patients with mild hepatic impairment. However, based on the observed increases in firsocostat exposure, dose adjustment should be considered for patients with moderate or severe hepatic impairment, and additional safety and efficacy data from future clinical trials will further inform dose adjustment.
