The Agricultural Microbiomes Thinkathon, held on 12 August 2023, in Denver, CO, U.S.A. brought together early-career and established scientists from academia, government, and industry to explore new approaches to microbiome research design, integration, and application. Organized by the U.S. National Science Foundation-funded Agricultural Microbiomes Research Coordination Network (AgMicrobiomes RCN), this community-based workshop emphasized collaborative thinking, interdisciplinary dialogue, and the cocreation of research ideas. Participants engaged in team-based activities aimed at transforming microbiome data into knowledge and actionable outcomes. Through structured breakout sessions, they developed research questions, interpreted microbiome datasets, and proposed projects focused on improving plant health and agricultural sustainability. These projects reflected systems thinking, practical applications, and shared interests in microbial function, plant–microbiome interactions, and data-informed management strategies. The Thinkathon demonstrated the value of workshop formats that move beyond passive or technical training to emphasize critical thinking, collaboration, and creativity. As microbiome data become more accessible and computational tools grow increasingly complex, the ability to pose thoughtful questions and connect research to real-world outcomes is essential. By combining active learning with interdisciplinary team science, the Thinkathon model provides a replicable framework for building capacity, empowering early-career scientists, and advancing innovation in agricultural microbiome research. Many bacterial traits important to host-microbe symbiosis are determined by genes carried on extrachromosomal replicons, such as plasmids, chromids, and integrative and conjugative elements. Multiple such replicons often coexist within a single cell and, due to horizontal mobility, have patterns of variation and evolutionary histories that are distinct from each other and from the bacterial chromosome. In nitrogen-fixing Rhizobium, genes carried on multiple plasmids make up a third of the genome, are necessary for the formation of symbiosis, and underlie bacterial traits, including host plant benefits. Thus, the genomics and transmission of plasmids in Rhizobium underlie the ecology and evolution of this important model symbiont. Here, we leverage a natural population of clover-associated Rhizobium in which partner quality has declined in response to long-term nitrogen fertilization. We use 62 novel, reference-quality genomes to characterize 256 replicons in the plasmidome and study their genomics and transmission patterns. We find that, of the four most frequent plasmid types, two (types II and III) have more stable size, larger core genomes, and track the chromosomal phylogeny (display more vertical transmission), while others (type I and type IV, or symbiosis plasmid, pSym) vary substantially in size and shared gene content and have phylogenies consistent with frequent horizontal transmission. We also find differentiation in pSym subtypes driven by long-term nitrogen fertilization. Our results highlight the variation in plasmid transmission dynamics within a single symbiont and implicate plasmid horizontal transmission in the rapid evolution of partner quality.IMPORTANCEUnderstanding how bacterial genes move through natural populations is critical for understanding how bacterial traits evolve. Nitrogen-fixing bacteria Rhizobium leguminosarum live in symbiosis with plants and are a model for studying plasmid transmission and how mobile genetic elements impact the evolution of bacteria and plants. Here, we characterize the genomes of a natural bacterial population, then use novel approaches to show that mechanisms of gene transmission vary across multiple plasmid types that coexist within R. leguminosarum cells. We find that changes in the frequency of specific pSym types are associated with the decline of symbiotic partner quality in strains isolated from environments undergoing long-term fertilization. These results underscore the importance of plasmid transmission and evolution in shaping ecosystem processes like nitrogen cycling via bacterial-plant symbiosis. Our study provides a framework for probing plasmid dynamics within natural bacterial populations and how plasmid transmission affects genetic diversity and ecological interactions in bacteria.

While loneliness profoundly affects mental health at every stage of life, adolescents today face unprecedented challenges finding meaningful social connections. Social media is a contributing factor that can improve the structure of relationships; however, function and quality are often impaired which significantly impacts connectedness. Lifestyle medicine clinicians can address the loneliness epidemic by promoting ways for adolescents to increase their connectedness to peers, school, and family. Specifically, clinicians can support the development of connectedness by recommending ways in which youth can build social-emotional skills. Using these concepts in all age groups is likely to be an important area for clinicians to focus with their patients.

ABSTRACT This paper refers to psychoanalytic challenges that the intense use of the internet presents in the treatment process of our adolescent patients. The use of the cyberspace allows to neglect physicality, so that hurtful developmental needs can be avoided or ignored. This may lead to various ways of emotional splits of body and mind. The two cases in the paper show different perspectives: For Mohammed, the internet is a temporary tool to which he sacrifices his bodily needs. John remains in the world of the cyberspace, as his relationships do not offer him support for his development.

Abstract Extreme cold events, which have become more frequent, can revert the direction of long-term responses to climate change. In 2021, record snowstorms swept the United States, causing wildlife die-offs that may have been associated with rapid natural selection. Our objective was to determine whether the snowstorms caused natural selection in Eastern Bluebirds (Sialia sialis). To test which mechanism most influenced their survival, we measured the morphology and coloration of fatalities and survivors at three sites. Survival was associated with a longer tarsus and with a wider, longer, and deeper beak, in support of the starvation and thermal endurance hypotheses. Additionally, bluebirds with more-ornamented plumage were less likely to have survived, perhaps because of an early energy investment in mate and site acquisition. As bluebirds encounter increasingly warm summer conditions, the longer extremities favored during the snowstorms may continue to be favored through their thermoregulatory benefits. However, the dull plumage coloration favored by natural selection during the snowstorms may be opposed by sexual selection benefits of more-ornamented plumage. Overall, responses to extreme events are difficult to predict from responses to long-term climate change, and responses to one event, such as the 2021 snowstorms, may not predict responses to a future extreme event.

Abstract Habitat loss is an important driver in the rapid decline of many insects. The restoration of roadside habitat has been touted as an opportunity to support communities of bumble bees and butterflies, two insect groups of conservation concern. However, it is unclear how current roadside restoration methods translate to pollinator abundance and diversity. Here, we ask how the plant communities of roadsides seeded with native and non‐native seed mixes affect bumble bee and butterfly communities 2–20 years after planting. We found that bumble bee and butterfly abundance and diversity were positively correlated with floral abundance and diversity. However, the pollinator community in plots planted with diverse native seed mixes did not differ from that found in plots seeded with species‐poor non‐native mixes, likely because many forbs in the native seed mixes had poor establishment, and all plots were readily colonised by species that were never planted. While plots with more native flowers did not necessarily support higher pollinator diversity, we did find several native plants that established well in roadsides and supported pollinators, including goldenrods (Solidago spp.), milkweeds (Asclepias spp.) and wild bergamot (Monarda fistulosa). Our data suggest that managing a more diverse and abundant floral community will benefit bumble bees and butterflies. Refining the composition of native seed mixes, adding more floral abundance to non‐native mixes and investing in management for native plant communities may increase the habitat value of roadsides for pollinator communities.

Chromosome number change is a driver of speciation in eukaryotic organisms. Carnivorous sundews in the plant genus Drosera L. exhibit single chromosome number variation both among and within species, especially in the Australian Drosera subg. Ergaleium D.C., potentially linked to atypical centromeres that span much of the length of the chromosomes. We critically reviewed literature on chromosome counts in Drosera, verified the taxonomy and quality of the original counts, and reconstructed dated phylogenies. We used the BiChrom model to test whether rates of single chromosome number increase, decrease, and chromosome number doubling differed between D. subg. Ergaleium and the other subgenera and between self-compatible and self-incompatible lineages. The best model for chromosome evolution among subgenera had equal rates of chromosome number doubling but higher rates of single chromosome number change in D. subg. Ergaleium than the other subgenera. Contrary to expectation, self-incompatible lineages had a significantly higher rate of single chromosome loss than self-compatible lineages. We found no evidence for an association between differences in single chromosome number changes and diploidization after polyploidy or centromere type. This study presents an exemplar for critically examining published cytological data and rigorously testing factors that may impact the rates of chromosome number evolution.

For patients on renal replacement therapy (RRT), "travel" and "independence" are rated as 2 of the top 5 factors that inform their choice of treatment modality. While home dialysis modalities offer patients a high degree of independence, the most common RRT in the United States is in-center hemodialysis (IHD). The limits imposed by IHD treatment can present a variety of challenges for patients who wish to travel. This study explored how IHD patients managed their travel and the role of dialysis social workers in executing travel arrangements for patients. We performed a qualitative descriptive investigation using semi-structured interviews with adults receiving IHD (n = 16) and renal social workers (n = 8) from Iowa, Minnesota, North Dakota, South Dakota, and Wisconsin. Data were analyzed using a constant comparative method. Three themes emerged from the interviews: travel process, travel-related barriers, and travel-related facilitators. The travel process entailed transient dialysis unit challenges and the need for multiple preparations and precautions. Barriers included comorbidities and not having a relationship with transient dialysis unit staff. Facilitators focused on the importance of travel and staff professionalism at transient dialysis units. Overall, there was lack of uniform protocols to guide the travel process at the patient and the dialysis unit levels. This study identified multiple perspectives regarding travel arrangements in chronic IHD patients. There is limited research on travel issues for IHD patients and this investigation is among the first to articulate barriers and facilitators associated with travel from the perspective of patients and social workers. Supporting travel for IHD patients can increase their sense of autonomy and provide opportunities to improve their quality of life.

This study investigates the role of pentraxin 3 (PTX3) in the regulation of inflammatory homeostasis involving anti-inflammatory miR-21 during lipopolysaccharide (LPS) stimulation in adipocytes. Using PTX3 knockout (PTX3 KO) mouse and primary stromal vascular cell models, this study determined the effect of PTX3 deficiency on the expression and secretion of miR-21 in brown adipose tissue (BAT) and brown adipocytes as well as the rescue effect of recombinant PTX3 on miR-21 during LPS-induced inflammation. Among three fat depots, BAT was the major tissue and fully differentiated brown adipocytes were the major cells that expressed miRNA-processing enzymes and produced miRNA. Moreover, brown adipocytes, but not stromal vascular cells, were the LPS-responsive cells in miR-21 production. PTX3 deficiency attenuated LPS-stimulated upregulation of miR-21 in sera and BAT. In wild-type brown adipocytes, LPS stimulation significantly upregulated cellular miR-21. Interestingly, this stimulatory effect of LPS was attenuated, and cellular and secreted miR-21 levels were reduced in PTX3 KO cells upon LPS stimulation. Treatment of recombinant PTX3 reversed the cellular and secreted levels of miR-21 and attenuated an LPS-stimulated increase in the expression of Tnf-α and Mcp1 genes in PTX3 KO adipocytes. PTX3 plays an anti-inflammatory role, in part through regulating miR-21 expression and secretion.
