We examined how resistance exercise (RE), cycling exercise and disuse atrophy affect myosin heavy chain (MyHC) protein fragmentation. The 1boutRE study involved younger men (n = 8; 5 ± 2 years of RE experience) performing a lower body RE bout with vastus lateralis (VL) biopsies being obtained prior to and acutely following exercise. With the 10weekRT study, VL biopsies were obtained in 36 younger adults before and 24 h after their first/naïve RE bout. Participants also engaged in 10 weeks of resistance training and donated VL biopsies before and 24 h after their last RE bout. VL biopsies were also examined in an acute cycling study (n = 7) and a study involving 2 weeks of leg immobilization (n = 20). In the 1boutRE study, fragmentation of all MyHC isoforms (MyHCTotal) increased 3 h post‐RE (∼200%, P = 0.018) and returned to pre‐exercise levels by 6 h post‐RE. Interestingly, a greater magnitude increase in MyHC type IIa versus I isoform fragmentation occurred 3 h post‐RE (8.6 ± 6.3‐fold vs. 2.1 ± 0.7‐fold, P = 0.018). In 10weekRT participants, the first/naïve and last RE bouts increased MyHCTotal fragmentation 24 h post‐RE (+65% and +36%, P < 0.001); however, the last RE bout response was attenuated compared to the first bout (P = 0.045). Although cycling exercise did not alter MyHCTotal fragmentation, ∼8% VL atrophy with 2 weeks of leg immobilization increased MyHCTotal fragmentation (∼108%, P < 0.001). Mechanistic C2C12 myotube experiments indicated that MyHCTotal fragmentation is likely due to calpain proteases. In summary, RE and disuse atrophy increase MyHC protein fragmentation. Research into how ageing and disease‐associated muscle atrophy affect these outcomes is needed.HighlightsWhat is the central question of this study?How different exercise stressors and disuse affect skeletal muscle myosin heavy chain fragmentation.What is the main finding and its importance?This investigation is the first to demonstrate that resistance exercise and disuse atrophy lead to skeletal muscle myosin heavy chain protein fragmentation in humans. Caffeine's metabolism is determined by CYP1A2 genotypes: AC/CC (SLOW) and AA (FAST). This trial evaluated CYP1A2 genotypes' impact on exercise and cognitive effects in 36 resistance-trained females assessed under placebo (PL) and caffeine (6 mg/kg bw anhydrous caffeine-CAF) conditions, before ingestion and throughout the session. 23andMe® (San Francisco, CA, USA) determined genotypes using saliva. Data were analyzed using two-way RMANOVA and paired-samples t-tests (p < 0.05). A significant main effect for genotype existed for leg press repetitions to failure (RTF) for CAF (p = 0.038), with the FAST group performing more repetitions than the SLOW (p = 0.027). There was a significant condition x genotype interaction for the subjective outcome index score (p = 0.045), with significant differences for time (p < 0.01) and between genotype (p < 0.001). Follow-up analysis revealed a higher total score (p = 0.028) following CAF for the FAST group and a lower total score (p < 0.01) in the SLOW group. Dizziness was reported following CAF in the SLOW group (p = 0.014; Cohen's d = 0.725). Aside from leg press RTF, subjective outcome index score, and dizziness, the genotype groups experienced similar responses to resistance exercise performance and subjective mood states following caffeine ingestion.

Background: Prophylactic lace-up braces are commonly used in athletics regardless of ankle injury history to reduce lateral ankle sprains (LAS). Objectives: The purpose of the study was to assess ankle and knee kinematics associated with chronic knee pathologies during a 90° cutting maneuver before and after a state of fatigue. Methods: Using a randomized crossover design, thirteen, active adults (6 males and 7 females) engaged in a 15-m beep test fatigue protocol with a 90° cutting maneuver test before and after the fatigue protocol, both with and without brace. Results: Brace condition significantly reduced Ankle Sagittal Displacement (ASD; 52°) compared to no brace condition (62°) (p .05). Peak knee flexion was significantly greater in brace (56°) compared to no brace condition (47°) and, likewise, took longer to reach it (10 ms) (p .05). Conclusions: Prophylactic ankle braces limiting ASD can cause the foot to land in a more dorsiflexed position at initial ground contact causing more rearfoot strike, which may contribute to chronic knee pathologies.

Softball is a sport that requires speed, agility, and lower-body power to be successful. Accordingly, strength and conditioning programs have focused on improving speed and lower-body power, which are beneficial to players performing softball-related tasks. The purpose of this study was to determine the correlation between absolute and relative lower-body strength to performance measures among female collegiate softball players. Archived data collected during pre-season performance testing for twenty-one (n = 21) NCAA Division II collegiate softball players was used for this analysis. Measurements included countermovement jump (CMJ), broad jump (BJ), linear speed (LS), 505 test for change of direction speed (COD), and shuttle runs. Absolute lower body strength (ALBS) was measured using a 3-repetition maximum hex-bar deadlift (HBDL) and body weight was used to calculate relative lower body strength (RLBS) of each player. Pearson's correlation coefficients were used to relate measures of lower body strength to each performance test. Significant (p ≤ 0.05) large to moderate correlations were discovered between RLBS and COD (505L: r = −0.59, 505R: r = −.63), 300 yard (yd) shuttle run time (trial 1: r = −0.47, trial 2: −0.50), and lower-body power (CMJ: r = 0.48, BJ: r = 0.52). ALBS was correlated only to the BJ (r = 0.43). The results suggest that relative strength is related more with COD, shuttle run, and lower-body power when compared to absolute strength. Therefore, strength and conditioning professionals should consider measures of RLBS when administering softball specific tests for developing and implementing a successful strength program in collegiate softball.

Chronic Thermogenic Dietary Supplement Consumption: Effects on Body Composition, Anthropometrics, and Metabolism
Multi-ingredient thermogenic supplements can acutely increase resting energy expenditure (REE) and subjective energy. However, less is understood about the effects of chronic consumption on body composition, metabolism, and subjective variables such as mood, sleep quality, and eating behaviors. Fifty-two healthy, exercise-trained participants (50% female; mean ± SD age: 23.5 ± 3.0 years; body fat percentage: 27.3 ± 8.0%) were randomized 2:2:1 to take a whey protein supplement alone (PRO; n = 20), in combination with a thermogenic supplement (PRO + FB; n = 19), or no supplement at all (CON; n = 13) for four weeks. Body composition, anthropometric, metabolic, hemodynamic, and subjective outcomes were collected before and after the intervention. Greater changes in REE occurred in PRO + FB as compared to CON (111.2 kcal/d, 95% CI 2.4 to 219.9 kcal/d, p = 0.04), without significant differences between PRO and CON (42.7 kcal/d, 95% CI −65.0 to 150.3 kcal/d, p = 0.61) or between PRO + FB and PRO (68.5 kcal/d, 95% CI −28.3, 165.3, p = 0.21). No changes in hemodynamic outcomes (blood pressure and heart rate) were observed. In exercising adults, four weeks of supplementation with protein and a multi-ingredient thermogenic product maintained fasted REE as compared to no supplementation, for which a decrease in REE was observed, without differential effects on body composition, anthropometrics, or subjective variables.
