ABSTRACT Cancer cell-autonomous type 1 interferon (IFN-1) production and signaling is frequently activated in response to DNA damage and has been associated with the development of therapy resistance in several cancer types. However, its cell-autonomous role in driving resistance in high-grade serous ovarian cancer (HGSOC), a disease defined by near-universal exposure to genotoxic therapy as frontline treatment, remains unclear. Specifically, whether IFN-1 functions in HGSOC as only a response to genotoxic stress or can independently act in driving resistance phenotypes has not been studied. Utilizing a syngeneic patient-derived model of cisplatin-sensitive (SE) and -resistant (CR) HGSOC, we demonstrate that chronic cisplatin exposure is associated with enrichment of IFN-1 signaling and the interferon-related DNA damage resistance signature (IRDS). Acute cisplatin treatment elicited dynamic, temporal IFN-1 signaling and responses in both sensitive and resistant cells, indicating a conserved stress response in resistant cells. Chronic, low-level exposure to exogenous IFNβ, in the absence of a DNA-damaging agent, was sufficient to phenocopy several features of chronic cisplatin driven resistance, including reduced therapeutic sensitivity, cell cycle arrest, and decreased proliferation. Notably, IFNβ driven resistance occurred without sustained IRDS or canonical interferon stimulated gene (ISG) induction, revealing alternative mechanisms for IFN-1 mediated therapy resistance. Together, these findings identify IFNβ as a functional driver of the development of resistance-associated phenotypes and highlight cell-autonomous IFN-1 signaling as a potential biomarker for resistance and a therapeutic target in platinum-resistant disease.

The research was conducted to establish the effectiveness of silver nanoparticles embedded in indium tin oxide (ITO) thin film conducting material in causing the textile azo dye effluent degradation. The percentages of azo dye degradation were as follows: 68.4%, 72.5% and 93.1% using electrocoagulation, ITO and ITO-AgNPs coupled electrodes, respectively. The results obtained from the two-factor ANOVA with replication indicated a significant difference (p = 0.0024) when the silver nanoparticles embedded on indium tin oxide electrodes were utilised in the degradation of these azo dyes in the effluent samples. The colour removal was 99.97% showing that the optimised process was effective and efficient in the degradation of these azo dyes in the textile wastewaters.

Fourier Transform is a low-cost method for improving duty cycle, resolving power, and signal-to-noise ratio in the Ion Mobility Spectrometry (IMS) experiment in a 2-gate IMS cell. By simultaneously pulsing both gates through a frequency sweep, the resulting data may be deconvoluted into a time-based mobility spectrum through a Fast Fourier Transform (FT). However, inconsistencies common in low-cost function generators result in spectral artifacts. In this work, an asynchronous stepped frequency FTIMS method is demonstrated, which uses a simple, software timed pulse generator compatible with any modern Analog-Digital Converter (ADC) system. By unlinking the frequency initiation and data collection using a long rise-time amplifier circuit, a stand-alone FTIMS with Faraday Plate detection was characterized in both single gate and FT modes of operation using the same IMS cell. Asynchronous stepped FTIMS parameters were investigated for system optimization with respect to resolving power, signal-to-noise ratio, and experimental time. Once optimized, asynchronous FTIMS demonstrated significant improvements in resolving power and signal-to-noise ratios without a significant increase in experimental time. By unlinking the frequency generation and data analysis, a simple Python script was demonstrated using a variety of commercially available ADC systems ranging in cost from several thousand to several hundred dollars (USD) without sacrificing spectral fidelity. A custom circuit was developed to allow a Raspberry Pi 4 Single Board Computer (SBC) to function as the data acquisition and control (DAC) interface for a low-cost stand-alone FTIMS solution.

Social science researchers and higher-education personnel have begun to recognize the unique and unequal experiences of first-generation, low-income, and working-class students, but what of those from such backgrounds that manage to reach the professoriate? What can we learn from their mobility experiences? What obstacles do they and others perceive standing in their way? Using original data on faculty and graduate students collected between 2018 and 2022, we analyze how diverse faculty (including graduate students) reflect on the relevance of social class background for upward mobility and success in academia. Results point to a shared understanding of ways in which cultural, social, and economic resources and resource constraints are influential although we also find noteworthy interpretational divergences by social location and identity. We discuss our most central results in these regards as well as their implications for building greater equity in higher education.

Purpose Drawing on Social Exchange Theory (SET), the purpose of this paper is to explore whether mentorship quality mediates the link between mentor functions (career support, psychosocial support and role modeling) and employee outcomes (organizational commitment, self-esteem and employee well-being). Design/methodology/approach Hypotheses were tested among 268 full-time employees who engaged in a mentoring relationship from a range of organizations using path analysis in AMOS. Findings Results showed that mentorship quality fully mediates the relationships between mentor functions and the employee outcomes of self-esteem and employee well-being. Originality/value This present study broadens the body of literature applying SET in workplace contexts and tests a model in which mentorship quality mediates the relationships between the mentor functions and protégé outcomes.

The dinuclear platinum(IV) compound {Pt(CH3)3}2(μ-I)2(μ-adenine) (abbreviated Pt2ad), obtained by treating cubic [PtIV(CH3)3(μ3-I)]4 with two equivalents of adenine, was isolated and structurally characterized by single crystal X-ray diffraction. The National Cancer Institute Developmental Therapeutics Program’s in vitro sulforhodamine B assays showed Pt2ad to be particularly cytotoxic against the central nervous system cancer cell line SF-539, and the human renal carcinoma cell line RXF-393. Furthermore, Pt2ad displayed some degree of cytotoxicity against non-small cell lung cancer (NCI-H522), colon cancer (HCC-2998, HCT-116, HT29, and SW-620), melanoma (LOX-IMVI, Malme-3M, M14, MDA-MB-435, SK-MEL-28, and UACC-62), ovarian cancer (OVCAR-5), renal carcinoma (A498), and triple negative breast cancer (BT-549, MDA-MB-231, and MDA-MB-468) cells. Although anticancer studies involving some adenine platinum(II) compounds have been reported, this study marks the first assessment of the anticancer activity of an adenine platinum(IV) complex.

ABSTRACT Disruptive journalistic innovation tends to challenge established news organizations and practices from the outside in and from the bottom up, especially when that innovation is driven by those considered on the periphery of accepted professional journalism. Journalists are skeptical about both those whom they perceive as “outsiders” and about potential threats caused by disruptive innovation. This study explores the journalistic discourse around four media innovations led by figures who defy clear classification into insider or outsider categories: CNN, USA Today, The Huffington Post and Semafor. Content analysis of coverage of the launch of these disruptors suggests greater acceptance of radically new media approaches when their purposes align with fundamental journalistic ideals. This U.S.-focused case study also contributes to the existing body of international research by examining how journalistic acceptance or rejection of entrepreneurial figures’ ventures relate to perceptions of those individuals’ belonging in the realm of journalism.
