A high-sensitivity and high-resolution brain PET system has been developed with a multiresolution detector design, using two LYSO crystal layers with different crystal sizes to achieve high time-of-flight (TOF) and depth-of-interaction (DOI) information simultaneously. In this study, we developed a list-mode ordered subset expectation maximization (OSEM) reconstruction algorithm incorporating TOF and DOI for this system. Then, we investigated the effects of TOF and DOI on the reconstructed image performance separately, using Monte Carlo simulations of a modified image-quality phantom and a Derenzo phantom that was placed off-center as well as in the center field of view. Finally, the system spatial resolution was assessed using reconstruction with both TOF and DOI. The modified image-quality phantom results showed that reconstruction with TOF could greatly reduce image artifacts, enhance background uniformity, and increase contrast recovery coefficients. The off-center Derenzo phantom results showed that reconstruction with DOI could greatly improve the tangential resolution. Using reconstruction with both TOF and DOI, <tex xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink">$1.25-\text{mm}$</tex> hot rods can be clearly identified. In conclusion, this work developed a list-mode OSEM reconstruction algorithm with incorporating TOF and DOI, achieving a <tex xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink">$1.25-\text{mm}$</tex> image resolution for this dual-layer brain PET system.
